Asthmatic Rats, 8 - Iso Prostaglandin F <sub> 2¦Á </ Sub> Levels And The Effect Of Intervention Experimental Study

Objective:Bronchial asthma is developed in childhood mostly,it has high incidence,but the pathogenesis is still unknown.Most researches show that the oxidative stress damage has close relation with asthma chronic airway inflammatory,also the 8-iso-PGF2αis a hotspot in the study of overoxygen damage.We established the rat asthma model,and take 8-iso-PGF2αas the main indicator,to discuss the influence of oxidative stress,overoxygen damage to asthma airway inflammatory and the significance in estimating the degree of asthma damage,the change of each indicators and pathology aider the anti-oxygen treatment and inhaling glucocorticoids,to provide experiment proofs in prevention and therapy of bronchial asthma.Methods:Fifty male Sprague-Dawley(SD)rats of 4 weeks old were divided into five groups randomly,10 as control group and 40 as Model, BUD,NAC and BUD+NAC group。The rats of control group were given intraperitoneal injection with 1ml 0.9%sodium chloride instead of 10% ovalbumin(OVA)in the sensitized stage,and 0.9%sodium chloride instead of OVA was inhaled through pump method in the challenge stage. The rats in other four group were given intraperitoneal injection with 1ml 10%OVA mixed with 10%aluminum hydroxide gel and the inactivated Bacillus pertussis on day 1,2%OVA was inhaled to sensitize the rats with pump methods for 7days two weeks later.The rat asthma model was established successfully.Three of the four groups of rat asthma model were treated with BUD,NAC and BUD+NAC.The rats were sacrificed 24h later aider the last challenge.Then the arterial blood,bronchial alveolar lavage fluids(BALF)and lung tissue of each group rats were taken,total cells number of BALF and the percentage of eosinophils (EOS)cells were detected,and the lung tissue histopathology was analyzed.We detected the expressions of 8-iso-PGF2αon plasma, 8-iso-PGF2αand IL-5 on BALF with ELISA,analysed the relativity among 8-iso-PGF2α,IL-5 and total cells number of BALF.Results:As compared with the other groups,the rats of asthma mode appeared allergic reaction signs similar as asthma patients,the lung histopathology showed typical airway inflammation,lots of inflammatory cells infiltrated the airway,bronchus tract narrowed,epithelia cells fall off,mucus excreted more,the total cells number and the differential cell counts including lymphocyte and eosinophils(EOS)in BALF were increased,also the expressions of 8-iso-PGF2αincreased on plasma and BALF(P<0.05).As compared with the asthma model group,allergic reaction signs and the degree of inflammatory cells infiltration on lung tissue histopathology were decreased in BUD,NAC and BUD+NAC group,the total cells number and the expressions of 8-iso-PGF2αand IL-5 were decreased,especially in the BUD+NAC group.Relativity analyse show that the content of 8-iso-PGF2αin plasma and BALF have positive relativity and have significant difference (P<0.05).The content of 8-iso-PGF2α,the total inflammatory cells number and the percentage of eosinophils(EOS)cells in BALF have positive relativity and have significant difference(P<0.01).Conclusion:We established the rat asthma model successfully,and found that the expressions of 8-iso-PGF2αon plasma and BALF were increased significantly.After treated with Budesonide and NAC,the content of 8-iso-PGF2α,the total inflammatory cells number and the percentage of eosinophils(EOS)cells in BALF were significant decreased and have positive relativity,which show that 8-iso-PGF2αcan reflect the degree of asthma airway inflammatory and estimate the effect of therapy. Budesonide may control the outbreak of asthma though reducing the inflammatory reaction,blocking the following oxygen damage. Anti-oxygen therapy(NAC,N-acetyl-L-cysteine)can affect the oxygen damage directly,alleviate asthma symptom,and co-work with Budesonide to improve the prognosis of asthma.
