Background and Objective:Reactive oxygen species are main free radicals in biosystem which is oxygen-rich functional group with active chemical property.It include superoxide(O2-),nitric oxide and hydroxyl radical(OH+),singlet oxygen,and other molecular species,such as hydrogen peroxide(H2O2)and peroxynitrite(ONOO-),causing harmful effect on most cells.Normally,there is dynamic balance to the generation and scavenging of ROS in the organism.When oxidative stress occurs,with the amount of ROS increasing and exceeding the ability of clearance,it will cause lipid peroxidation,DNA oxidative damage,protein abnormal expression,resulting in detrimental effect on organism.ROS has played a vital role in the development of many myocardial injuried disease,such as heart muscle ischemia/reperfusion injury,heart failure,hypertension.Hydrogen dioxide,one form of ROS,can initiate strong lipid peroxidation,react with any cell,resulting in myocardial cell apoptosis and functional disorder.Curcumine is phenolic pigment derived from the root of Curcuma longa L. one of ginger family plant.It can be widely used in food additive and dye for its stable character and low toxicity.Multitude reaseach prove curcumine has a series of pharmacologic action such as anti-inflammation,anti-atherosclerosis, anti-tumor,anti-oxidative effects on the basis of protecting cells from adverse factors.Curcumine,as a kind of oxygen free radical scavenger,can inhibit evidently active oxygen like superoxide,hydrogen peroxide,nitrite group.It also has powerful antagonistic effect on injury induced by H2O2 in human fibroblasts and enthodial cells.It protects heart muscle in rats from ischemia by relieving ischemic change in electrocardiogram induced by isoprenaline,and improving cell action in oxygen deficit situations.It is supposed that the clearance of free radicals accounts for the mechanism.By far,it is not demonstrated that the role and mechanism of curcumine on myocardial injury in cell level in vitro.This reseach supply experimental evidence to prevent and cure cardiovascular disease for curcumine,through culturing cardiomyocytes of neonatal rats in vitro,establishing myocardial damaged model by H2O2 and observing the effect of oxidative damage on myocardial cells of neonatal rats.Method:Culture the myocardial cells of neonatal rats,then divide into five groups randomly,control group,H2O2 group,different concentration of curcumine groups.Assay the viability of myocardial cells using MTT;The activity of dehydrogenase(LDH),The content of malonaldehyde(MDA)and surperoxide dismutase(SOD)were measured by their respective kits;Flow cytometry detect the rate of apoptosis.Result:Compared as control group,the cell viability decrease in H2O2 group.Except the low dose group of curcumine,the other curcumine groups significantly increased the viability of myocardial cells.The optical density value in myocardial cells increase with the raise of the concentration of curcumine.All curcumine groups inhibited release of LDH,decreased the content of MDA and increased SOD activity of myocardial cells injured by H2O2.Compared with H2O2 group,curcumine with 50μmol/L concentration decreased the rate of apoptosis evidently.Conclusion:Curcumine could obviously protect cultured neonatal cardiomyocytes from H2O2 damage,and its protective effects is connected with concentration of curcumine.The underlying mechanism may be result from its antioxidant properties.
