Objective:In the last few years, human Y chromosome polymorphisms are increasingly interested by scientists, not only for population genetics or evolutionary studies but also for forensic application.More and more anthropologists study human migration and evolution by comparing the genetic distance and clustering different populations using Y-chromosomal polymorphic genetic marker. The considerable interest is the potential ability of Y-chromosome analysis to unravel the race and ethnic background of a given DNA profile. The results of this study preserved and enriched south Chinese Han ethnical gene informational resources.Method:We co-amplified DYS19,DYS389Ⅰ,DYS389Ⅱ,DYS390,DYS391,DYS392, DYS393,DYS385a,b,DYS437,DYS438,DYS4391 STR loci and 92R7,M103,M110,M111, M117,M119,M120,M122,M128,M130,M134,M139,M159,M15,M168,M175, M176,M178,M17,M214,M217,M231,M3,M45,MS,MS0,M60,M7,MS,M88, M89,M9,M91,M95,M98,RPS4Y,SRY1532 SNP markers from YCC.We investigated 50 unrelated male individuals of Southern Han individuals.Result:Among the 50 individuals,48 haplotypes were detected. For STR markers,47 alleles and 24 haplotypes were detected, with allele frequency and gene diversity ranging from 0.02-0.68, 0.4775-0.9526.18 haplotypes were detected in 38 SNP loci. Conclusion:The result showed that the studied Northen Han in China while indicated significantly differences with Southern Han.In order to study the allelic frequency distribution and haplotype diversity of Y-chromosomal STR and SNP loci of Chinese southern population, evaluate their usefulness in forensic paternity testing and human identification, and carry out Chinese population genetic analysis.
