Objective:According to the pathogenesis of Vascular dementia in both Chinese medicine and medical sciences,as well as Rhodiola herb research progress,explores Rhodiola herb to improve the VD animal model study and the memory ability curative effect,expounded the partial possible action mechanism,to be for the purpose of for this medicine preventing and controlling VD to provide the experiment to rest on.Materials and Methods:1.Reference Etsuko Miyamoto,Thomas and other documents of bilateral carotid artery ligation before permanent brain ischemia induced vascular dementia model.2.Reference to the literature using random number table this 24 mice were randomly divided into three groups,namely,Chinese crude drug(Rhodiola herb) group,western medicine (Nimodipine) group,blank model group,setting up a sham group.3.Delivery methods:Oral gavage administration,delivery volume for 1ml/100g weight.For 28 days after administration, While the rats in sham operation control group and model group were given DW of the same dosage.4.Used Morris water maze test devices and dynamic image acquisition and analysis system for learning and memory testing,It was adopted to test the rats' memory change of each group(1months after the occlusion of the CCA).The escape incubation period expressed the rats' memory ability.Various parameters recorded by the computer records.5.HE staining of pathological changes of the hippocampus;Immunohistochemical detection of rat hippocampal CA1 NR1 expression of Bcl-2,Bax protein expression was observed.6.Measured brain NOS absorbance of brain tissue NOS activity was by spectrophotometry,calculates the brain to organize the NOS vigor.Results:1.Water maze training from the first day onwards,the model group than the sham-operated group escape latency significantly longer(P<0.05),4-5 days more obvious differences(P<0.01).And the treatment group first two days of the incubation period was shorter than evade model group(P<0.05),from the first four days after the differences in the performance of two relatively stable,but also significant(P<0.05).Sham-operated group compared with the escape latency there is no significant difference(P>0.05).At the same time, four rats with the increase in the number of training,spatial memory capacity and search platform to shorten the time.2.Treated rats than the model group and the choice of routes campaign platform found significantly improve the efficiency of the model platform at the time, the effective area of distance than the control group at different levels have decreased,but also significant(P<0.05).3.Sham group had no obvious pathological changes.Model group cortex, the hippocampus and capsule loose structure,a softening see larger lesions,accompanied by glial cells and vascular proliferation;gray see a part of ischemic neuronal morphological changes,cell body shrinking space around the halo;hippocampal CA1 cells are disordered,soma significantly smaller nuclear pyknosis,nucleolus disappeared,dense cytoplasm.Small treatment group occasionally softened cortex lesions,soma narrowed,but the shape and close to normal,more than the number of models;hippocampal CA1 region of the cell body size and normal close.4. Model group NR1 expression weakened,compared with the treatment group was significant(P<0.05);model of dementia compared with the sham-operated model group NOS activity increased significantly with a significant(P<0.05);model Rhodiola herb and western medicine group and the comparison group NOS activity increased significantly(P<0.05).5.Bcl-2 and Bax in the sham-operated group hippocampal CA1 a small amount of expression.VD model group coloring positive cells increased,the number increased significantly,which Bax changes more significantly(P<0.05),the ratio of bcl-2/Bax compared with the control group decreased significantly.Treatment group bcl-2 and Bax-positive cells also increased,by deep coloring, especially bcl-2 change significantly,the ratio of bcl-2/Bax significantly higher than the control group,but also significant(P<0.05).Conclusions:1.Rat bilateral carotid artery ligation caused chronic cerebral ischemia can be a better simulation of vascular dementia,is an ideal model of VD.2.VD model for the Rhodiola herb,can improve the level of knowledge and pathological changes,improve learning and memory abilities.3.Rhodiola herb can reduce the VD in the hippocampus of rats NR1 expression,EAA reduce toxicity,neurons play a protective role.4.VD Rhodiola herb can reduce brain tissue of rats NOS activity and reduce NO production.5.VD rat hippocampus Rhodiola herb regulation of Bcl-2 and Bax protein expression and inhibit neuronal apoptosis,protect nerve cells,and promote the recovery of brain function.
