Influenza is the acute infection disease of respiratory passage caused by influenza virus. It's jeopardizing to human health and safe,to economic development and so on.Till now ,there has some medicine to resist flu virus,such as ion channel blocker,neuramidinase inhibitor,etc.Those medicine has some impairment in somewhere,but Chinese medica has advantage.The pharmacodynamics studies are to confirm the effectiveness of Dureping injection(DRP) on influenza viral FM1 infected mice. Immune regulation studies and cytokine studies on macrophage invivo and invitro are to explore the indication of DRP of reducing immunologic injury on influenza viral FM1 infected mice.The experiment includes two parts:The first part, is focus on the function of macrophage of mice infected influenza viral FM1,study about the effection on content of NO,interleukin-8 (IL-8),tumor necrosis factor-α(TNF-α) and intercellular adhesion molecule-1(ICAM-1) in supernatant of macrophage of mice (Ana-1) infected influenza viral FM1.The second part,is concentrated on the effection of DRP to the lungs of mice infected influenza viral FM1. Study about the effection on content and expression of interleukin-8 (IL-8) ,tumor necrosis factor-α(TNF-α) and intercellular adhesion molecule-1(ICAM-1) in homogenate of lungs of mice infected influenza viral FM1.Results of Part One:1 Effect of Dureping injection on content of NO in supernatant of macrophage of mice (Ana-1) infected influenza viral FM1 in vitro:Compared with model group, the content of NO was increasing both in SHL control group and DRP treated group(P﹤0.05), large dose group of DRP had the lowest content, it was as same as the content in SHL control group,and the medium dosage group was the next in order. The tendency had visible time-effect relationship. The effect became obviously with the extension of time.2 Effect of Dureping injection on content of TNF-αin supernatant of macrophage of mice (Ana-1) infected influenza viral FM1:Experimental result showed that, compared with model group, the content of TNF-αwas lower both in SHL control group and DRP treated group(P﹤0.05), and SHL had more therapeutic efficacy(P﹤0.001). Large dose group of DRP had the lowest content of TNF-α(P﹤0.001), and the medium dosage group was the next in order, the minute dose group had the weakest effect. Large dose group of DRP had the lowest content of IL-8, and the medium dosage group was the next in order, the minute dose group had the weakest effect., while the effect became obviously with the extension of time.Results of Part Two:1 Effect of Dureping injection on lung index in mice infected influenza viral FM1: Arising from experimental results, lung index in model group markedly increased in contrast with sham group(P﹤0.0001). Compared with model group, the lung index was lower in all the other groups, there was a remarkable difference(P﹤0.05 ~ 0.0001), the DRPz -treated group was the best one among them.2 Effect of Dureping injection on content of TNF-α,IL-8,ICAM-1 in lung of mice infected influenza viral FM1:The content of TNF-α,IL-8,ICAM-1 in lung homogenate of mice in model group were increased,there was a significant difference in contrast with sham group(P﹤0.001). All the NRP-treated group could reduce content of TNF-α,IL-8,ICAM-1 (P﹤0.05 ~ 0.01). Compared with model group, there was a remarkable difference(P﹤0.01)3 Effect of Dureping injection on mRNA transcriptional level of TNF-α,IL-8,ICAM-1 in lung of mice infected influenza viral FM1:The mRNA transcriptional level of TNF-α,IL-8,ICAM-1 in lung of mice in model group were increased,there was a significant difference in contrast with sham group(P﹤0.01). All the NRP-treated group could reduce the mRNA transcriptional level of TNF-α,IL-8,ICAM-1 (P﹤0.05 ~ 0.01). Compared with model group, there was a remarkable difference(P﹤0.05).
