Objective: IgAN is the primary giomerular disease, which is the most common type of pathology. To establish the improved IgAN rat model, observe-the effect of the model and explore the improved IgAN mechanism.Method: To choose SPF 25 male Wistar rats'were randomly divided into : the normal group,model group, Jinshuiqing early treatment group, the Jinshuiqing late treatment group and the positive control group (xueniaoan group) five groups, through oral bovine serum albumin (BSA) and intravenous staphylococcal enterotoxin B (SEB) establishment of rats IgAN model, the basis of this to be added to environment,dietary and biological factors infection pathogens causing compound with damp-heat syndrome performance of the model. The early group were treated from 7th week, once a day, to 12th week; The late group and xueniaoan group from 9th week respectively, once a day, to 12th weekend. The animals were killed to observations : 1. general state, 2. microscopic examination detected in urine RBC, biuret protein in urine was detected quantitatively 3. automatic biochemical analyzer detection of renal function (BUN using enzymatic assay, creatinine used picric acid); 4. RIA serum levels of IL-6, Immunohistochemical detection of renal fabrice TGFβ1 content. 5. Optical microscopy and fluorescence microscopy to observe animal model of renal pathology.Results: 1. General state: The normal group from the start of the experiment to an end in spirit, and reactive activities, eating, without obvious abnormalities; model group rats had varying degrees of tired, fluffy hair, eating few, short-urine, stool soft rot andsome appeared edema, anus swelling, etc damp-heat syndrome performance and the treatment group were treated and the performance improved significantly.2. After detection of RBC and proteins in urine:the normal group has not appeared hematuria and proteinuria, the treatment group blunt and significantly reduced compared proteinuria with the model group was statistically significannot (P<0.01) The morning group and the late group, xueniaoan group was statistically significant (P<0.05) .The late group and xueniaoan group was not significant (P>0.05). 3. Renal function (blood urea nitrogen, creatinine) testing, the treatment groups urea nitrogen, creatinine decreased significantly. Compared with the control group compared with statistical significance (P<0.01) the early group and the late group. xueniaoan group was statistically significant (P<0.05) .The late group and xueniaoan group was not significant (P>0.05).4. IL-6 levels in the show that the treatment groups were significantly lower. Compared with the control group was statistically significant (P<0.01) .Detect by image analysis demonstrates that the mean of OD value group with the early group, xueniaoan group was not significant (P>0.05), The late group and xueniaoan group was not significant (P>0.05).5. Light microscopy showed normal mesangial cells, the capillary loop open. glomerular atrophy tubular structure of the basic rules of stromal hyperplasia and infiltrat ion of inflammatory cells. Fluorescence microscopy revealed normal glomerular mesangial area are weak or non-IgA, IgG deposition; the treatment groupsglomerular and tubular IgA IgG showed granular yellow-green fluoresce, the early group was lightest.Conlusion: The fine can improve IgA modified rat model of damp-heat symptoms, reduce urinary protein and urine RBC, kidney reduce immune complexes in renal tissue deposition, and through reduced IL-6 and TGFβ1 expression, Inhibition of glomerular mesangial cell proliferation and extracellular matrix accumulation and reduce glomerular and tubular-interstitial injury, thereby improving renal function, the treatment of hematuria, and delaying the process of renal fibrosis effective herbs, but confirmed Qingrelishi method in children hematuda the importance of early treatment, it is worth further study and clinical application.
