Objective: Our research was designed to found the renal injury model caused by Aristolochia manshuriensis Kom on rats, and to explore the pathogenesis of its renal injuries. The preventive and therapeutic effects of Chinese Caterpillar fungus mycelium powder on the renal injuries caused by Aristolochia manshuriensis Kom were also studied.Methods: 40 healthy wistar male rats were evenly randomized distributed into four groups. Group 1 were normal controls, Group 2 and group 3 were aristolochic acid treated group and Aristolochia manshuriensis Kom decoction treated group. Group 4 were treated with Aristolochia manshuriensis Kom decoction and Chinese caterpillar fungus mycelium powder. Aristolochia manshuriensis Kom decoction 10g/(kg·d) for lavage was used to found the renal injury model caused by Aristolochia manshuriensis Kom on rats. 8 weeks later, the growth rate of body mass, quantitation of 24 hours urine protein and urine N-Acetyl-β-D-glucosaminidase (NAG) were determined. Nephridial tissue paraffin section were made to observe the structure of nephridial tissue under the light microscope and tubulointerstitial pathological scores were computed. The expressions of transforming growth factorβ1 were tested in each group by immunohistochemical methods.Results: 8 weeks later, the growth rate of body mass significantly decreased in the other groups than in group 1. Quantitation of 24 hours urine in group 3 and 4 were higher than group 1 (P<0.05), so were that in group 2(P<0.01). NAG significantly increased in the other groups than in group 1. NAG in Group 4 were lower than group 2 and group 3(P<0.01), and that of group 2 were higher than group 3(P<0.01).Tubulointerstitial pathological scores in group 2 were higher than group 3(P<0.05). The scores of group 4 were lower than group 3, but the differences had no statistical significance (P>0.05). TGF-β1 were found highly expressed in the other groups than in group 1(P<0.01). Expressions of TGF-β1 were higher in Group 2 and 3 than that in group 4(P<0.01), and that in group 2 were higher than that in group 3(P<0.01).Conclusion: (1)Aristolochia manshuriensis Kom decoction 10g/(kg·d) for lavage can successfully found the renal injury model caused by Aristolochia manshuriensis Kom on rats. It will cause the tubulointerstitial fibrosis. (2) Aristolochia manshuriensis Kom decoction and aristolochic acid A will result in the overexpression of TGF-β1, which participate in the formation of tubulointerstitial fibrosis. (3) Compared with group 2 and group 3, Chinese Caterpillar fungus mycelium powder can decrease urine protein and urine NAG, alleviate the renal pathological injuries, down-regulate the fibrosis cytokine. It has distinguished preventive and therapeutic effects on the renal injuries caused by Aristolochia manshuriensis Kom.Our research suggests that Chinese Caterpillar fungus mycelium powder may has preventive and therapeutic effects on the renal injury caused by Aristolochia manshuriensis Kom and provide an important proof for preventing and treating this kind of renal injuries.
