Human papillomaviruses (HPV) are small DNA virues, which characteristically infect squmous epithelial cells of the skin and mucosa. As a sexual transmitted disease, HPV infection is characterized by a very high rate of infection rate. It is generally accepted that persistent HPV infection is the most important factor in the development of condyloma acuminate and cervical cancer. But there are still many open questions about the association between HPV and Bowenoid Papulosis. A large number of studies demonstrated that HPV E6 and E7 proteins could interact with host genes and affect the cell cycle in different degree, interfere the normal cell proliferation and differentiation and ultimately result in benign and malignant proliferative lesions of the infected epithelial cells. At present, it is not completely undersdood how HPV leads to dysregulation of the cell cycle. In this study, we try to detect and type HPV in condyloma acuminate and Bowenoid Papulosis; then we detect the expression of HPV 16 L1, p53 and CD1a, investigate their changes and significance.At first, we amplied and typed HPV DNA with the consensus primer MY09/11 by PCR- RFLP in 52 paraffin-embedded biopsy specimens from condyloma acuminate and Bowenoid Papulosis lesions. Then we examined the expression of HPV16 L1, p53 and CDla by immunohistochemistry technique in 40 samples, the results are as fellows:1. HPV DNA is detected positive in 18/20 cases of condyloma acuminate, with the majority of cases for HPV type 6 and 11.2. HPV DNA is positive in 17(53.1%) of 32 cases of Bowenoid Papulosis, 14 cases of them are HPV16, one of them is HPV18, one of them is HPV32 ,another is both HPV6 and HPV16.3. The expression of HPV 16 L1 in condyloma acuminatum and Bowenoid Papulosis is 80% and 90% respectively. There is no expression in normal tissue.4. The expression of p53 in condyloma acuminatum and Bowenoid Papulosis is 30% and 45% respectively. There is no expression in normal tissue. 5. We find a significant reduction in the density of CDla positive cells in condyloma acuminatum and Bowenoid Papulosis compared to normal tissue.The results indicate that condyloma acuminata mainly arises from HPV6 infection, secondly from HPV11; HPV16 infection is strongly associated with the pathogenesis of Bowenoid Papulosis.HPV16 L1 has no specificity, there are cross reactions between defferent types. p53 protein might play role in the pathogenesis of condyloma acuminatum and Bowenoid Papulosis. A significant reduction in the density of CDla positive cells in condyloma acuminatum and Bowenoid Papulosis suggests that HPV infect derease Langerhans cells, which may play important role in HPV's escape of immunologic surveillance.
