Grape Seed Extract From Prostate Cancer Pc-3 Cells The Role Of Experimental Research

Grape seed extract (GSE) has shown the pleiotropic anticancer effects on differentcancer cells.Objective: (1) To study the effect of GSE on proliferation and apoptosis of PC-3 cells. (2)To analyze,the pathway in which GSE induce apoptosis of PC-3 cells. (3) To observe theenhanced response of PC-3 cells to radiation.Methords: (1) MTT assay was applied to test the inhibitory effects of GSE onproliferation, of PC-3-cells. (2) Invert microscopy and-transmission electron microscopy(TEM) were applied to observe the morphologic changes in PC-3 cells. (3) PI staining,AO/EB staining and Annexin V-FITC/PI were applied to study the effects of GSE onapoptosis of PC-3 cells. (4) Western blot was used to study the pathway of apoptosis. (5)Flow cytometry was applied to study the enhanced response of PC-3 cells to radiation.Results: (1) The proliferation of PC-3 cells was inhibited in a dose- and time- dependentmanner by MTT assay. (2) Changes in PC-3 cells morphology were observed by invertmicroscopy and TEM. (3) PI staining assay indicated that GSE could arrest PC-3 cells inG0/G1 phase. Annexin V-FITC/PI showed that GSE could induce apoptosis. (4) Westernblot showed that GSE induced apoptosis in the caspase-3 pathway. (5) Annexin V-FITC/PIassay showed that the response of PC-3 cells to radiation was enhanced by GSE. Besides,GSE could change the arrested phase of PC-3 cells after be radiated.Conclusion: The paper studied the effects of GSE on prostate cancer PC-3 cells in thelevel of cell and sub-cell. We testified for the first time that GSE was cytotoxicity to PC-3cells. All the results showed that GSE could induce the apoptosis and necrosis ofandrogen-independent prostate cancer PC-3 cells in the mitochondrion-dependent manner.And GSE could enhance the response of PC-3 cells to radiation.
