Aim:This study was conducted to explore the cooperatively protective effect of Adenosine and L-Arginine on ischemia-reperfusion injury in rat myocardium and to illustrate its mechanism.Methods:The hearts of rats were used to made ischemia-reperfusion model in our experiment.All hearts were subject to 30 minutes local ischemia,then were reperfused for 60 minutes,fifty Wistar rats were divided into 5 groups randomly,Each group had 10 rats:①Sham operated group:only wear the line and do not ligature the coronary artery;②Model matched control group:only complete the model operation;③Adenosine group:complete the model operation,1 minute before ischemic to infuse the Adenosine(2mg/kg)in tail vein;④L-Arginine group:complete the model operation,15 minute before reperfusion to infuse the L-Arginine(100mg/kg)in tail vein;⑤Adenosine+L-Arginine group:complete the model operation,1 minute before ischemic to infuse the Adenosine(2mg/kg)in tail vein,and 15 minute before reperfusion to infuse the L-Arginine(100mg/kg)in tail vein.Observed indexes include:the monitor of the electrocardiogram;②CK and LDH in blood;③myocardial SOD and MDA;④myocardial ultrastructure observation.Results:①L-Arginine and Adenosine+L-Arginine could reduce the incidence of reperfusion arrhythmias.Compared with model matched control group,the incidence of reperfusion arrhythmias of Adenosine were not significantly different.Compared with L-Arginine,Adenosine+L-Arginine could reduce the i nc i dence of reperfusion arrhythmias better.Adenosine,L-Arginine and Adenosine+L-Arginine all could make the raising S-T segment of electrocardiogram lower and Adenosine+L-Arginine was significantly better than either Adenosine or L-Arginine;②By improving the ability to anti-oxidize,Adenosine, L-Arginine and Adenosine+L-Arginine all could prevent the release of intracellular enzyme and reduce the ischemia-reperfusion injury,and the result of protection in Adenosine+L-Arginine was significantly better than to give Adenosine or L-Arginine singly;③Myocardial electron microscope observation showed that myocardial ultrastructure of Adenosine,L-Arginine and Adenosine+L-Arginine were better than that of model matched control group,and among them Adenosine+L-Arginine was the best of all.Conclusions:①Adenosine has protective effect on ischemia-reperfusion injury;②L-Argin ine has protective effect on ischemia-reperfusion injury;③Adenosine+L-Arginine has cooperatively protective effect on ischemia-reperfusion injury.
