PartⅠThe Expression and Location of HCK Gene During the Development of Rattus HeartObjective To investigate the mRNA expression of HCK (hemopoietic cell kinase) gene in the developing rattus heart and to analyze the relationship between HCK gene expression and the heart development.Methods Rattus hearts in embryonic days 12, 15, 19 were included in this study. The mRNA level of HCK gene expression was detected by the methods of RT-PCR and in situ hybridization, and the results were analyzed by the statistical methods. Results In the developing rattus hearts, HCK gene was mainly expressed in the interstitial tissue, but no expression in the cadiocyte. The levels of HCK gene mRNA expression were 0.19±0.09, 0.38±0.07, 0.59±0.10 alternatively, and it was up-regulated with the embryonic aging. Conclusion HCK was expressed in the interstitial tissue of the developing rattus hearts, but no expression in the cadiocyte. With the development of rat hearts, the expresion of HCK gene in the level of mRNA is dynamic. HCK may play a role in the heart development. PartⅡExpression of IFRD1 gene in the Developing Rattus HeartObjective To investigate the expression of IFRD1 (interferon-related developmental regulator 1) gene during the development of Rattus heart.Methods Rattus hearts in embryonic days 12, 15, 19 (E12, E15, E19) were included. The expression of IFRD1 gene was detected by RT-PCR methods, the expression of IFRD1 protein weas detected by Western Blot and Immunohistochemistry methods. Results (1) IFRD1 gene and protein were expressed in Rattus hearts of E12, E15 and E19. The expression level was decreased during the development of Rattus heart. (2) IFRD1 protein was mainly expressed in the atria, ventricles and trabecular muscles, while no expression in atrial-ventricular valves, outflow tracks and large arteries. Conclusion IFRD1 gene was down-regulated during the development of Rattus heart and mainly expressed in the atria, ventricles and trabecular muscles. IFRD1 may play a role in the heart development.
