Traditional Chinese drug is our traditional weapon of curing disease, it Contains great vitality, has been applied to the medical treatment for thousands of years. The study of isolating natural drugs' component, purfying natural drugs, mechanism of action have developed for many years. The medicinal chemistry has been attentive by scholars of chemistry and biology. The alkaloids is an important bioactive substance, Has significant biological activity. Recently, with the discovery of bacteriostasic, anticancer, analgesia, anaesthesia and decompression activity of alkaloids, etc, the medicinal chemistry research of alkaloids is accelerating. Systematically studying the chemical activity and biological activity of Active ingredients of traditional Chinese drugs and deeply developing new drugs is becoming the more and more scholars' topic of concern.In this paper, the bacteriostasic activity of Active components of Chinese medicine was studied by microcalorimetry, the thermal power-time curves of the bacteriostasic activity of alkaloids were determined by TAM Air, then analyzed the thermal power-time curves, constructed the theoretical model and thermodynamic equation of bacterium growth, and calculated related thermodynamic functions, systematically studied the thermodynamic characters. The study will provide theoretical basis for guiding the synthesis of new specific medicine.1 The chief content 1.1 The extraction and characterization of drugsThe effective components of the seven kind of drugs were extracted and purified by vacuum evaporation, soaking, distillation, filtration, extraction, precipitation, Recrystallization, etc. Then the extracts were characterized by using the methods of infrared spectroscopy, the melting point, thin layer chromatography, liquid chromatography-mass spectrometry, the results show that the pure extractions have berberine, isotetrandrine, palmatine, nuciferine, caffeine, ursolic acid.1.2 The study of bacterial growth metabolism in different concentration of drugsIn order to find out the regulation of bacteriostasic metabolization and the optimum concentration of drugs, the power-time curves of bacteria in different drugs were determined at five kind of temperature, then the curves were analyzed by using logistic equation so that the growth rate constant of bacteria in drugs was obtained at different concentration, the relationship between growth rate constant and concentration, the optimum concentration was determined. The results show that all the seven Active ingredients have Inhibition to E.coli, S.aureus, B.subtilis, drug concentration and growth rate constant of bacteria have a kind of linear relationship, the Inhibitory effect of the seven active ingredients can be got by contrast of the optiumum concentration of drugs, and find out the relationship of growth rate constant and temperature, then the optimum temperature was established.1.3 The study of the growth of bacteria under the influence of drugs by usingtransition state theory model.The thermodynamic functions in activation state were calculated, from the perspective of thermodynamics to find out the law of the growth of bacteria at different alkaloids and concentration and temperature, the law is that ΔG has little change with the change of temperature, the numbers of ΔG are between 82 and 84 KJ/moL; at the same temperature the numbers of ΔS gradually increase with the increase of concentration of the drugs, from negative to positive number. The magnitude of the equilibrium constant of bacteria in activated state is same, which is 10-15, the equilibrium constant decreases gradually at the same temperature with the increase of concentration of the drug, when the temperature is between 25 and 37℃, the equilibrium constant increases gradually at the same concentration of the drug with the increase of temperature, and the optimum allowable concentration and the optimum allowable temperature of each alkaloid were calculated, the all data are consistent with the data got by analyzingthe curves of Pt-t.2 The main innovative points in the thesis2.1 The crystals of alkaloid and saponin which were extracted from traditional Chinese drug were characterized by using the methods of infrared spectroscopy, the melting point, thin layer chromatography, liquid chromatography-mass spectrometry to determine the structure and purity of the extraction, also study the same type of alkaloids that have the common mother structure with different functional Groups, and the relationship of structure and nature was established.2.2 Thermodynamic functions in Activation state were calculated by the study of activated state properties of bacteria under the influence of drugs at different temperature and concentration, from the perspective of thermodynamics to find out the law of the growth of bacteria at different alkaloids and concentration and temperature, the optimum allowable concentration and the optimum allowable temperature of each alkaloid was calculated by using these thermodynamic functions in Activation state, the result is consistent with that got by analyzing the curves of Pt-t.
