Puerariae Radix, the root of Pueraria lobata{Willd.) Ohwi and Pthomsonii Benth., is a very important Chinese traditional medicine recorded in the Pharmacopecia of People's Republic of China (ed. 2000). It is a commonly used drug for antipyretic, spasmolytic and antihypertensive therapy . The author has investingated the chemical constitutes, fingerprints, and its quality determination.In this thesis, the chemical constituents of the ethanol extracts from Pueraria lobata were studied, 16 compounds were isolated with silica gel, polyamide, preparative TLC, preparative HPLC etc. on the basis of analysis of NMR, MS data and measuring of physicochemical characters, their chemical structures were elucidated as puerarin (1), daidzein (2) , daidzin (3) , 3'-hydroxy puerarin (4) , 3'-methoxy puerarin (5) , 3'-Methoxydaidzein(6), 5-hydroxyl ononin(7), formononetin (8), ononin (9), genistein (10), genistin(11), daidzein 7,4'-diglucoside (12), 8-C-apiosyl(1â†'6) glucoside of daidzein (13), 4-hydroxy-3-methoxy cinnamic acid (14), β -sitosterol(15), daucosterol (16), respectively. Of all these compounds, compound 6, 14 were isolated for the first time from Pueraria,. While 7 was isolated from Pueraria lobata(Willd.) Ohwi for the first time.To establish the analysis method of Radix Puerariae, 13 batches of Pthomsonii Benth. and 4 batches of Pueraria lobata(Willd.) fingerprints were studied by means of " The Similarity Calculation Soft of Fingerprint Chromatography of Traditional Chinese Medicine". The thesis also establishes the fingerprint for the herbs, and the peaks have been assigned.The quality determination method for the herbs was established. |
