To establish the Laboratory Mouse Frozen Embryos Archieve included three parts: select viable methods for cryopreservation of mouse embryos, control the embryos quality by genetic monitoring and set up the database management system.1. To optimize empirically the conditions of superovulation in our laboratory, we allocate the C57BL/6 mouse and 129/J mouse into three groups: 2.5IU group was given 2.5IU dose of PMSG and HCG, 5.0IU group was given 5.0IU dose of PMSG and HCG, 7.5IU group was give 7.5IU dose of PMSG and HCG. The result shows that the total number of zygotes isolated (C57BL/6 mouse) of 5.0IU group is significantly higher than that of 2.5IU group and 7.5IU group (P<0.05). The total number of zygotes isolated (129/J mouse) of 7.5IU group is significantly higher than that of 2.5IU group and 5.0IU group (P<0.01).To choose some suitable medium for the culture of mouse embryos, we evaluated the blastocyst formation of mouse embryos in M16 medium, CZB medium, and KSOM medium (5% CO2, 37℃). The result shows that the blastocyst formation rate in KSOM is significantly higher than that in M16 (P<0.05) and CZB (P<0.01), the result in M16 is not significantly different from that in CZB (P>0.05).To study the effects of cryopreservation on the development of...
