objective To explore the relationship between P38 mitogen-activated protein kinase (P38MAPK) and nitric synthase (iNOS) in the rat kidney during the late phase of ischemic preconditioning (IPC),aiming at clearifing the possible mechanism of the delay renal protection in ischemic preconditioning .Metheds 60 male wistar were randomly divided into 5 groups (n=12): sham operation group; ischemic reperfusion(IR)group; ischemic preconditioning + ischemic reperfusion(IPC+IR) group; SB203580 group; AG group.Each group was subdivided into two groups (n=6) after 24h;48h of ischemic reperfusion respectively. The renal function was examined,the renal pathological morphology was observed by HE and the cell apoptotic was estimated with TUNEL, at the same time the expression of P38MAPK and iNOS protein in the renal tissue were assayed by western blotting.Result The values of serum creatinine were lower in IPC+IR group than IR group (p<0.05);the lowest values appeared 48h in IPC+IR group. The expression of P38MAPK and iNOS protein were all observed in the sham operation group;IR group;IPC group,while markedly increased expression appeared 48h in IPC+IR group;.There was no P38MAPK expression in the SB203580 group; at the same time we have found that the iNOS expression in the SB203580 group was less than IPC+IR group but more than the IR group;In the AG group,the protein of the iNOS was not found, and the P38MAPK expression was not inflenced by AG..Conclusion P38MAPK and iNOS proteins are both involved in the delay renal protection of IPC,also P38MAPK is the up stream substance of iNOS protein.
