Objective To provide the evidence for the application of DYS460 (A7.1) in forensic science and population genetics by investigating its polymorphism in the Han population lived in Shanxi province.Method To collect respectively 100 unrelated males' venous blood 500ul, who live in Shanxi province. Different tissues of one corpse were analyzed including blood, muscle, heart, liver et al. 20 father-offspring pairs were investigated. The blood of 20 females were collected All blood-specimens were anticoagulanted with EDTA, the Genomic DNA was extracted using the method of phenol-chloroform after ingested by proteinase K and amplified the DYS460 locus by using PCR. The PCR amplification products were separated on 8% non-denatured polyacrylamide gels electrophoresis and associated with silver stained. A direct calculation and the x~2 test were applied to our data.Result There are total 4 alleles in DYS460 site in Shanxi Han population, called DYS460*9, DYS460*l0, DYS460*11, DYS460*12, the GD(Gene Diversity ) value is 0.7214, the frequencies of alleles are from 0.12-0.35. There are not amplified products among females. The typing results of different tissues from one individual were identical. The amplified products of female's samples were negative. Mutations were not found from 15 father-offspring pairs. Conclusion It shows the DYS460 in Shanxi Han population is high valuable in the application of forensic Medicine and population genetics.Conclusion It shows the DYS460 in Shanxi Han population is high valuable in the application of forensic Medicine and population genetics.
