| Objective:Observe the effect of the Chinese herbal medicine to the gene expression of DM rats' Collagen mRNA of type IV and some other indexes. Inquiry into the mechanism of the compound formula of the prevention and cure of the Diabetic nephropathy (DN) in their earlier period. Methods:30 male Wistar rats, four-month old, 200 ± 20g , were randomly divided into four groups. They are normal group, DM group, western medicine control group(Ruitai), Chinese herbal medicine control group, all the rats in DM group were not given food for 10 hours before being injected, STZ (53mg/kg) was injected one time to the abdomen, thus they become the DM models. After eight weeks' Chinese herbal medicine treatment, compared with Ruitai, Observe: (1) micro blood sugar (2) Ualb of 24hours (3) the expression of β 1mRNA, the expression of Collagen mRNA of type IV (4) the change of kidneys' condition in each group. Result:(1)The micro blood sugar of the DM rats, Ualb of 24hours, the expression of the kidney transforming growth factor β 1mRNA and the expression of Collagen mRNA of type IV have risen bviously. (2)Compared with the DM group, the blood sugar, the Ualb of 24hours, TGF- β 1mRNA and the expression of Collagen mRNA of type IV have all dropped. (3)The DM rats TGF- β 1mRNA and the expression of Collagen mRNA of type IV have direct proportion, the same with TGF- β 1mRNA and Ualb. (4)We can see from observing the kidneys' condition that: the kidneys' structures are intact in the normal group, the kidneys' sizes didn' tincrease; the velamen cells of the DM rats have got hyperplasia, the velamen areas have widened, the basic substance has got hyperplasia, GBM has thickened diffusely , the cells of the kidneys' upper skin has swollen, parts of the kidneys' capillary vessels have expanded, the red cells have filled up and are in a line; the upper pathology changes of both the Chinese medicine control group and the western medicine control group are... |
