The Clinical Observation Of Chinese Herbal Compound In PV Patients And Study On The Expression Of JAK2Gene MRNA In HEL Cell Line By Using Realgar

Objective:1.To observe the therapeutic effect of Shuganhuoxuetang formula intreatment of polycythemia vera (PV),and to test hemoglobin counts,with the aim to providebasis for the mechanism of Chinese herb and clinical treatment.2.To research the cellproliferatin and expression of JAK2gene influence when different concentrationas ofrealgar acts on the HEL cell lines with mutation of JAK2V617F,in order to provide the PVa new treatment.Methods:20patients with PV were randomly divided into two groups.The TCM andWestern medicine therapy group(10cases) was treated with Shuganhuoxuetang formulaand the western medicine control group(10cases) was treated with Hydroxyurea andinterferon medicines for3months of therapy respectively.The clinical effect were observedduring the course;hemoglobin counts.Using MTT method to confirmed the exact role ofrealgar inhibit proliferation of HEL cell line.To determine the mRNA expressions of JAK2in HEL cell line after using different concentrations of realgar by RT-PCR assay.Results:The total effective rate of the TCM and Western medicine therapy groupreaches90.00％,can effectively reduce hemoglobin count of PV patients,while that of thewestern medicine control group reaches80.00%,which shows no significant differencebetween the two groups(P>0.05).there is significant difference between both groups aftertreatment on traditional Chinese medicine symptoms(P<0.05),this is to say,The TCM andWestern medicine therapy group is better than the western medicine control group toimprove the quality of life of patients.The onset time of clinical efficacy in both groupswith significantly difference (P<0.05),and there is also very significant difference betweenboth groups on onset time of clinical efficacy of traditional Chinese medicine symptoms(P<0.01).Both groups can decrease the level of the blood hemoglobin counteffectively(P<0.01),and the decreasing effect of treatment group is the same with that ofcontrol group(P>0.05).Realgar inhibit the proliferation of HEL cells with obvious time-anddose effects.The mRNA relative expressions of JAK2are significantly different amongControl group、25mg/L doses realgar group、50mg/L realgar group and100mg/L dosesrealgar group(P<0.01);25mg/L doses realgar group、50mg/L realgar group and100mg/Lrealgar group can lower the mRNA relative expressions of JAK2(P<0.01);The mRNArelative expressions of JAK2in100mg/L realgar group are lower than50mg/L realgargroup (P<0.01);The mRNA relative expressions of JAK2in50mg/L realgar group arelower than25mg/L realgar group (P<0.01).Conclusion:Shuganhuoxuetang formula has affirmatory curative effect in thetreatment of PV, with the ability of decreasing the hemoglobin count effectively,and beeffective in preventing the occurrence of clinical symptoms,such as hemorrhage andthrombosis,improve quality of life and prognosis.The effect of realgar’ inhibiting HEL cellproliferation is related with its lowering the mRNA expressions of JAK2.
