That means the material must not elicit an unresolved inflammatory response nor demonstrate extreme immunogenicity of cytotoxicity. The cytotoxicity test is the most important one in the biocompatible system. MTT test is a routine test method giving reproducible and quantitative results allowing the evaluation of significance. In addition, it will not fall in the controversy of using animals. Now MTT is popular when evaluate biocompatibility of dental materials.The objective of the first part is to evaluate the cytotoxicity of a new kind of orthodontic wire and bracket on human gingival fibroblast (HGF) in vitro. The method is to determine light absorption value of every hole after 24h , 48h and 72h of contact of the colored and unstained orthodontic bracket and wire with HGF cells by MTT assay, and calculate the relative multiplication rate of cells, and evaluate them by six degree tests for cytotoxicity. The results are that the cells in two infusion which has different concentrations respectively multiplicate greatly and toxicity ranges from 0-1. There was no significant difference of the two kinds of material (p>0.05). We have the conclusion that the colored orthodontic wire and bracket have good biocompatibility.With the increasing popularity of Flow Cytometry, more and more investigators prefer to have their cytotoxicity test with FCM expecting to gain more information basing on DNA level.
