Object To explore the effect of Terfenadine on guinea-pig'electrocardiogram and on preventing and curing ventricular arrhythmia.Method To record the guinea-pig'electrocardiogram after giving terfenadine by intraperitoneal injection, and to analyze the variation of heart rate and QTc interval. To observe wether terfenadine would effect the model by giving different concentration of BaCl2. Amiodarone was given as positive control group.Resultâ‘ Different concentration of terfenadine-treated (0.54,1.08,2.16and 4.32mg/ml)can all induce the abnormality of guinea-pig'electrocardiogram- stepping down heart rate after 40 minutes,and have significantly statistic difference comparing with NS ,DMSO control group and prodosage (P<0.05) excluding 0.54,1.08mg/ml of terfenadine. Different concentration of amiodarone-treated(7.2and10.8mg/ml) can also induce the abnormality of guinea-pig'electrocardiogram- stepping down heart rate after 40 minutes, and have significantly statistic difference comparing with NS control group and prodosage (P<0.05). excluding 3.6mg/ml of amiodarone .Different concentration of terfenadine-treated (0.54,1.08,2.16and 4.32mg/ml) can all induce the abnormality of guinea-pig'electrocardiogram- prolonging QTc interval after 40 minutes, and have significantly statistic difference comparing with NS ,DMSO control group and prodosage (P<0.05) Different concentration of amiodarone-treated(3.6,7.2and10.8mg/ml) can also induce the abnormality of guinea-pig'electrocardiogram- prolonging QTc interval after 40 minutes, and have significantly statistic difference comparing with NS control group and prodosage (P<0.05). In our experiment dose extent,the time to prolong QTc interval to maximal degree is uniform and the prolonging degree have significantly dose-dependent in both medicine after giving terfenadine or amiodarone.â‘¡0.54mg/ml of terfenadine can significantly decurtate the duration of ventricular arrhythmia induced by 0.3%BaCl2 comparing with NS control group(P<0.05),it can not significantly decurtate the duration of ventricular arrhythmia induced by 0.5%,0.7%BaCl2 comparing with NS control group(P>0.05). Different concentration of terfenadine-treated(1.08,2.16and4.32mg/ml) can significantly decurtate the duration of ventricular arrhythmia induced by 0.3%BaCl2 comparing with NS control group(P<0.05), and there are not statistical significance in group comparision and comparing with 0.54mg/ml of terfenadine. Different concentration of terfenadine-treated(1.08,2.16and4.32mg/ml) can significantly decurtate the duration of ventricular arrhythmia induced by 0.5%,0.7%BaCl2 comparing with NS control group and 0.54mg/ml of terfenadine(P<0.05), and there are not statistical significance in group comparision.
