At present, drug-resistance has been a worldwide problem. Pathogens which have resistance to antibiotics emerged widely in the hospitals and communities. Because the generation of drug-resistance has many factors, multi-resistant organisms have emerged and diffused rapidly. The research of drug-resistance has become the focus in the world because the generation and diffusion of drug-resistance bacteria are rapidly. The Chinese drugs showed their powerfully vital force in resisting drug-resistance for their special theories and remarkable clinical curative effects. The components of Chinese drugs are complex. Bacterial resistance to them is not easily generated. And many Chinese drugs have excellently inhibitory activity.In the paper, drug-resistance of E.coli to four kinds of Chinese drugs was studied by microcalorimetry. The difference of drug-resistance of E.coli to the different active components of Chinese drugs was compared.This paper includes six chapters.Chapter one: The first chapter summarizes research status about drug-resistance and Chinese drugs, development and actuality of thermokinetics and microcalorimetry, and introduces the chief contents and study significances of the paper.Chapter two: The Study of drug Resistance of E.coli to Active Components of Magnolia officinalis by Microcalorimetric Method. The magnolol and the total extraction of Magnolia officinalis were extracted and isolated. The drug Resistance of E.coli to magnolol and the total extraction of Magnolia officinalis was studied. And the difference between them was compared. The results showed that the inhibitory activity of the monomer was better, but the drug resistance of E.coli to it was produced more rapidly.Chapter three: The Study of drug Resistance of E.coli to Active Components of Phellodendron amurese Rupr by Microcalorimetric Method. The berberine and the total alkaloids of Phellodendron amurese Rupr were extracted and isolated. The drug Resistance of E.coli to them was studied. And the difference between them was compared. The results showed that the inhibitory activity of the monomer was better, but the drug resistance of E.coli to it was produced more rapidly.Chapter four: The Study of drug Resistance of E.coli to Stephenia tetrandra S. Moore by Microcalorimetric Method. The tetrandrine and the total alkaloids ofStephenia tetrandra S. Moore were extracted and isolated. The drug Resistance of E.coli to them was studied. And the difference between them was compared. The results showed that the inhibitory activity of the monomer was better, but the drug resistance of E.coli to it was produced more rapidly.Chapter five: The Study of drug Resistance of E.coli to Sinomenium acutum Rehd. et Whils. by Microcalorimetric Method. The sinomenine and the total alkaloids of Sinomenium acutum Rehd. et Whils. were extracted and isolated. The drug Resistance of E.coli to them was studied. And the difference between them was compared. The results showed that the inhibitory activity of the total alkaloids was better, and the drug resistance of E.coli to it was produced more slowly.Chapter six: The Study of Cross Resistance and Multidrug Resistance. The cross resistance and multidrug resistance of E.coli to the active components of the four Chinese drugs were studied.There are many researches on the drug resistance of bacteria to antibiotics. But the researches on the active components of Chinese drugs are few. In this paper, the monomers and the total extractions were extracted from the four Chinese drugs. And the monomers were characterized by X-ray diffractometer and HPLC/MS/MS. Then the drug resistance of E.coli to the monomers and the total extractions was studied. Some important parameters were obtained, which can provide academic bases for clinic treatment. Microcalorimetry as a new method in studying drug resistance can measure correctly MIC. And the microcalorimetric method has the merits of rapid measurement, quantitative determination, reliable results, good repeatability.
