ObjectiveCoronary artery disease (CAD) is one of the major diseases threatening the health of the mankind and the disease incidence is upgrading and the age of onset becomes younger and younger. Arthrosclerosis (As) is the pathological base of CAD and the prevention and cure are paid close attention on. As is the chronic pathological procedure and the oxidative stress plays an important role on its forming. The major pathogenic factor is oxidized low density lipoprotein (ox-LDL) and the injury of vascular endothelial cell for the oxidative stress. After adhere to EC, monocytes transfer into the foam cell, then form the early process of As. Clinical and experimental study indicate, JIANGZHINING has better effects on accommodating blood ester and anti-oxidation. So we choose the vivo model with ApoE gene deficiency mouse and the vitro model with monocyte to study JIANGZHNING's influence on the forming of As and anti-oxidation, then approach the mechanism of anti-oxidation and provide the experimental basis for the effects of various effective ingredients, the functional pathway and the prevention and cure of As with the herb.Methods1. ApoE gene deficiency mice, aged 8 weeks, weighed 16±2g, were randomized into JJIANGZHINING high dose group, JIANGZHININGlow dose group, atorvastatin calcium positive control group and model group. 12 weeks later, blood was taken from the heart of the mice for determining the biochemical indicator of serum; aortic root was taken for pathological dying pathological image analysis;2. Serum containing JIANGZHINING was prepared with Wistar rats, then sterilized by filtering;3. the functional pathway through direct anti-oxidative experiment of LDL oxidizing into ox-LDL was approached in vitro;4. The effect of the serum containing JIANGZHINING on the expression of CD36 WAS detected in monocyte/macrophage with flow cytometry;5. The effect of serum containing JIANGZHINING was detected on monocyte/macrophage swallowing ox-LDL.Results1. Compared with the model group, both JIANGZHINING high and low dose group can markedly decrease the level of total cholesterol (TC), triglyceride (TG) of ApoE gene deficiency mice ( p<0.05).
