Gastric carcinoma is a most common malicious cancer all over the world, which severely threatens human being's health. It is especially rife in Japan, China, Chile, Iceland, Austria, Finland and Hungary. In our country, there are about 200 thousand new increasing gastric carcinoma cases every year, which is 17.2 percent of all the malignancy. This occupies the first place in the incidence of all malign tumors. The death rate of gastric cancer in China is 23.02 percent of all the malignancy death rate and become the first in different kinds of cancer mortality. In recent 30 years, the incidence and mortality rate of gastric carcinoma of some developed countries have obviously decreased, which the rate of that in China still keeps high. Naturally it greatly attracts the researchers' attention. Gastric cancer is caused by the mutual influence of dozens even hundreds of genes and their protein, which interact with each other and form a complex relationship. Although they function differently, they often get the cell's proliferation out of control through changing the expression level. Consequently this kind of gene becomes an important object in current research of gastric carcinoma. The research offers theoretical evidence to make clear the pathology of stomach cancer and to help the basic research and clinic therapy of gastric carcinoma. It is of significant importance to choose gene expression as the starting point and use the gene chip to study mRNA level's expression variance of gastric carcinoma tissue and normal tissue, In this article, we hybridized the gene chips with more than 14 thousand known human genes and tagged EST to fluorescenced cRNA derived from the gastric carcinoma and normal tissue. Then we transferred the fluorescenced signals to digital signals in order to compare the differences of gene expression between gastric carcinoma tissue and normal tissue through the analysis of the statistics. To have an accurate result, we used the method of Real Time RT-PCR to choose some cases from the result and check them. After strict selections, we found that 34 genes were up-regulateddramatically and 35 were down-regulated in the gastric carcinoma tissue, which showed the close relation between them and gastric carcinoma. Among all the genes, the expression variances of steapl, aurora-a, sfrp2, p-catenin> duspl and pou6f2 were very obvious and therefore they are worth researching. But nowadays few researches have been done on it. "With the help of some documents, I present this thesis to interpret and predict their structures and functions.
