Abstract/Summary:
BackgroundEarly detection and treatment of colorectal cancer can improve long-term survival and quality of life of patients.However,due to early colorectal cancer has no obvious,specific clinical symptoms or signs,early diagnosis and treatment of the disease is still difficult.Although a variety of biomarkers associated with colorectal cancer have been found so far,they are still difficult to be used for diagnosis and targeted therapy.In addition,the high mortality of colorectal cancer is mainly due to tumor recurrence and metastasis after surgery.Therefore,in-depth study of the molecular mechanism of colorectal cancer occurrence,invasion and metastasis is conducive to early detection of colorectal cancer and targeted therapy of colorectal cancer,thereby enhancing the survival rate and quality of life of patients with colorectal cancer.The study found FOXK1 can regulate embryonic development,maintain the balance of metabolism,and it is closely associated with occurrence and development of colorectal cancer.Experimental studies showed that down-regulation of FOXK1 can inhibit the proliferation,invasion and metastasis of colorectal cancer cells.We studied the effect of down-regulation of FOXK1 in SW480 on the expression profile of this sort of colorectal cancer cell line by microarray technology,aiming to shed some light on the oncogenic mechanism of FOXK1 and discover new therapeutic targets to treat colorectal cancer.MethodsAccording to our previous experimental results,we selected SW480 cell line to conduct experiments and grouped them into control group and experimental group respectively.The experimental group was transfected with FOXK1-siRNA and the control group was transfected with Scrambled-siRNA.Targeted knockdown of FOXK1 in colorectal cancer cell lines was performed,and verified the expression of FOXK1 after transfection by Western blotting.The RNA was analyzed by Nanodrop2000 ultramicro spectrophotometer and Agilent 2100 Bioanalyzer to control the quality of subsequent studies.We investigated the effect of down-regulation of FOXK1 on expression profile of colorectal cancer cell line SW480 by microarray technology,genes with |Fold Change|>2 was considered upregulated or downregulated,P<0.05 was considered significant.ResultsThe study showed that 888 genes were differentially expressed in SW480 cells transfected with siRNA-FOXKl,of which 491 genes were up-regulated and 397 genes were down-regulated.ConclusionThe results of our study indicated that many of differentially expressed genes were reported to be involved in tumor development.FOXK1 may regulate the occurrence and development of colorectal cancer by multiple signaling pathways.
