1,3-oxathiolane nucleoside analogues are useful antiviral agents, especially effective against DNA or RNA viruses.In this paper, the synthesis of two compounds of these analogues, i.e. Lamivudine and Emtricitabine, was studied. Both of the drugs are anti-HBV and anti-HIV agents, approved by FDA in U.S.A. in 1995 and in 2003, respectively.On the basis of reported synthetic routes, this paper achieved some improvements on the reaction steps and reaction conditions to reduce the byproducts. A new recrystallization method for the products was adopted.All of the intermediates have been confirmed by 1HNMR, and the aimed products by MS, 1HNMR, and elemental analyses. |
