The mechanism of encephalopathy is one of modern medical research hot spots.Among them,imflammation of cerebral ischemia becomes an important reseach domain,and as a pathological link of theinflammary injury, blood brain barrier(BBB) affects the cascade damage after cerebral ischemia. seriously.Treatment of Chainese medicine expresses good effect to many kinds of brains source disease.Especially,itis remarkable to the curative effect on cerebral ischemia. Its cilinicsuperiority is dominant.In the paper itputs in tumor necrosis factor alpha( TNF-a) which produces in the procedure of cerebral ischemia-reperfusion injury,blocks TNF-a in blood circulation with mono-antibody of TNF-a,and discusses thefunction changing of BBB and the mechanism of cerebral ischemia –reperfusion.This paper includes two parts: documental research and experimental research..The documental research discusses modern mechanism study on permeability of bood brain barrier aftercerebral ischemia reperftsion.The experimental research includes three parts:The first part studys on influence of increased permeability of boood brain barrier after cerebral ischemiareperftsion by blocking tumor necrosis factor alpha( TNF-a), and discusses protection effect ofQingkailing on hydrocephalus introduced after cerebral ischemia reperftsion. Adult male SD rats receiveddouble common carotid clamping. These rats divide into 9 groups randomly: normal group, 24h-modelgroup, 72h-model groups, 24h-treatment gruop(Qingkailing), 72h-treatment group (Qingkailing), 24hanti-TNF-α group, 72h anti-TNF-α group, 24h-treantment group (Qingkailing+anti-TNF-α), 72h-treatmentgroup (Qingkailing+anti-TNF-α). We obeserve change of cerebral containing water, aquaporin-4expression and albumin of brain capillary vessel., and study effect of anti-TNF-α and Qingkailing.The second part studys on changes of related cytokine expression after cerebral ischemia reperftsion byblocking tumor necrosis factor alpha (TNF-a), and effect of Qingkailing. Model and animal are same asPart1. Indexes include serum TNF-a conten, serum sICAM-1 content, serum P-selectin content and braintissue ICAM-1 expression. It studys on changes of related cytokine expression after cerebral ischemiareperftsion, and Disint oxicatingeffect of Qingkailing .The third part studys on changes of BBB morph after cerebral ischemia reperftsion by blocking tumornecrosis factor alpha (TNF-a), and effect of Qingkailing on these changes. Model and animal are same asPart1. We observe BBB morph change of paraffin section HE staining. We expect to reveal the effects ofTNF-a and Qingkailing on BBB morph after cerebral ischemia reperftsion.The results showed:1. Blocking TNF-α and using Qingkailing can reduce cerebral edema after ischemia reperftsion1.1 Blocking TNF-α and using Qingkailing can decrease cerebral water contentAfter 24h an 72h of cerebral ischemia reperftsion, Qingkailing reduces cerebral edema, With application ofanti-TNF-α, cerebral water content is no obvisously different from norma brain tissue. This indicate thatanti-TNF-α can make a recovery of damaged brain tissue. So TNF-α acts a very important roll onmechanism of ischemia reperftsion.1.2 Blocking TNF-α and using Qingkailing can improve expression of AQP4.After 24h an 72h of cerebral ischemia reperftsion, Qingkailing increases AQP4 expression of brain tissue.After 24h, this effect is more manifest. It shows Qingkailing can recovery AQP4's function after injury,and makes water back to blood circulation. This effect of Qingkailing becomes more better with more time.Contrasting to normal brain tissue, the groups of applying anti-TNF-α do not recovery. But contrasting tomodel, the AQP4 expression of these groups all increase after 24h and 72h.Meanwhile, the treatments group of application with anti-TNF-αhave a more powerful expression of AQP4than no application. This indicates that integration of Chinese medicine and anti-TNF-α is a good waywhich have more excellent, more overall, more safe effect.1.3 Blocking TNF-α and using Qingkailing can decr...
