Diabetese Mellitus (DM) is a metobalic disease,resulted from thedefective insulin secretion,which is charactered byhyperglycemia.Chronic hyperglycemia usually induces the seculardamage,inadequacy and exhaustion of all sorts of viscera,especially theeye,kidney,nerve and cardiovisceral vessel. Type 1 diabetes is a kind of organ-specific autoimmuneendocrinopathy,which occurs in pancreatic beta cells.At present,thecure of type 1 diabetes mostly depends on injectinginsulin,however,injecting insulin chronically will induce manycomplications. Chinese herbs have mild side-effect,and its therapeuticaleffectismild,persistent and comprehensive.In order to search effectiveherbs which can cure type 1 diabetes,we perform the followingexperiments:1,Establishing mice models of DM;2,Validating the effectof decreasing blood glucose of six herbs;3 , Investigating thetherapeutic mechaniam of effective hurbs which can decrease bloodglucose level.1 Establishing mice models of DM and its pathological process.1.1 The method of establishing mice models: In this study,diabetes mellitus was induced in C57BL/6J by themultiple low-dose streptozotocin(STZ) (intraperitoneal injection ofSTZ at a concentration of 40mg/kg body weight per day for fiveconsecutive days).1.2 The pathological process of mice models (1) The body weight of diabetic mice is significantly lower thanthat of control mice,and its consumption of food and water is muchhigher than that of control mice. (2) The blood glucose levels of diabetic mice were significantlyhigher than those of control mice from day 6,and received the standardof DM model(≥16.7mmol/L),and the blood glucose levels lasted untilthe end of experiment(day 30). (3) The morphological results showed diabetic pancreatic isletsto be extensively infiltrated with lymphocyte,whereas pancreaticislets from control mice display normal structure.(4) The serum C-peptide levels from DM mice were much lower thannomal mice,and its GADA and IAA levels were much higher than those ofcontrol mice. From these results,it can be concluded that mice models characteredwith autoimmune disease preferably simulate type 1 diabetes.2 Screening effective hurbs which can decrease glucose levels We chose 6 herbs(Ginseng,Bitter melon,Tartary buckwheat,GiantKnotweed Rhizome,Kudzuvine Root,White Mulberry Root-bark) which havebeen reported as being efficient in decreasing blood glucose levels,andobserved their effects of decreasing blood glucose levels.We found thatGinseng could reduce the blood glucose in diabetic mice.3 Effects of Ginseng on diabetic mice3.1 Ginseng could reduce the body weight of diabetic mice,and increase the food and water intake of them;3.2 Ginseng could reduce the lymphocytes infiltration of diabetic mice;3.3 The immunological function of Ginseng on diabetic mice: (1) Ginseng could increase the serum C-peptide levels of diabeticmice,and reduce GADA and IAA levels of them; (2) Ginseng could reduce the expansion of spleen and thymus indiabetic mice; (3) Ginseng could restrain the DTH reaction induced bydinitrofluorobenzene(DNFB) in diabetic mice. In conclusion,these observations reveal Ginseng might treat DMby reducing the damage of autoimmunization on diabetic mice.
