Puhuang is a common used Chinese herb with long history in traditionalChinese medicine,which is the pollen of Typha angustifolia L.,T. oriential Presland the other plants of the genus Typha .Firstly it is included in Shen Nong'sCannon of Materia Medica. It is sweet in taste and neutral in nature. Itstherapeutic action is related to the channels of the liver and heart-sac. It has theeffects of cooling blood to arrest bleeding and promoting blood circulation toremove blood stasis, Usually,there are the raw drug and carbonized productused in clinic.The raw drug, which has the effects of promoting bloodcirculation, dispering blood stasis and relieving pains,it is often prescribed totreat dysmenorrheal, amenorrhea,mass in the abdomen,postpartum lochiorrhaeand pain caused by ecchymoma and traumatism. The carbonized product, whichhas the effects of arresting bleeding, is effective for various kinds of internal andexternal hemorrhage. Puhuang was processed far from Song Dynasty, rifely carbonized from QingDynasty. Carbonizing can strengthen the effect of arresting bleeding accordingthe traditional processing theory. So, on the basis of inheriting traditiongalexperiences,using modern chemical isolating and analytical methods, combinedwith the examination of pharmacological activity,the prospect will be broad forus to study carbonized puhuang. Three sections are included in this dissertation with the purpose of researchon the chemical components after carbonizing puhuang.Section of document Being the first part, there is a documental review of the scientific research onpuhuang including documentary references, chemical constituents,pharmacological effects, clinical application and processing case.Section of experimentPART ONE Eight components were isolated from the chromatography process of the70% alcohol extraction from the carbonized puhuang. Using the spectrumtechnology such as HNMR,13CNMR,IR and EI-MS, 5 structures were identified as the 1following: β-sistosterol, n-hexacosane, daucosterol, hentriacontanol-6n-heptatetracone and 1 mixture of saturated fatty acids which main containlignoceric acid and docosanoic acid.PART TWO By comparing the extract contents of the carbonized product and the rawdrug, the contents of water and 70%alcohol extracts from the raw drug weremore than that from the carbonized product. On the contrary, the content of etherextact from the carbonized product was more than that from the raw drug. Theresults indicated carbonizing could decompose the chemical components intosome fatty ones, leaded to the increasing of the content of the ether extract andthe decreasing of the contents of water and 70%alcohol extracts . Determinated the contents of the total flavonoids, using the 0.1mol/LAlCl3colorimetry with typhaneoside as the evaluation index, The calibration curve arelinear at ranges of 0.0062~0.0310mg/mL(r=0.9997,n=5). The contents of thetotal flavonoids in the carbonized product were 0.29%, those of the totalflavonoids in the raw drug were 1.84%. Determinated the contents of the total polysaccharide, using4%phenol-H2SO4 colorimetry with glucopyranose as the evaluation index, Thecalibration curve are linear at ranges of 0.0048~0.0288mg/mL(r=0.9996,n=6).The contents of the total polysaccharide in the carbonized product were 0.23%,those of the total polysaccharide in the raw drug were 2.44%. Determinated the contents of tannins by KMnO4 titration, the contents oftannins in the carbonized product were 3.69%, those of tannins in the raw drugwere 2.53%. The petroleum ether extracts from the raw drug and the carbonized productwere analyzed by GC-MS after saponificated and methanolized, and theirconstituents were searched through MAINLIB and REPLIB. As a result: Eitherin the raw drug or in the carbonized product, 26 components were foundrespectively, which had 20 same components and 6 different ones.
