| 1. Objective and Background Crohn's Disease(CD) is a chronic, recurrent and nonspecific inflammationthat can attack any place on whole intestinal tract, especially in distal endof ileum, proximal end of colon and crissum, and also can attacks many placesat the same time. Such symptoms as bellyache and diarrhea are its main symptomsin clinic, complicated by abdominal mass, fistulization and other symptoms inexterior intestinal tract and in body. Although its pathogenesy is not known,some factors such as surroundings, microbes, inheritance and immunifaction havealready been found to be relevant to CD according to research reports,especially the immunifaction becomes focus. And CD is supposed that the suscwith its inheritance were attacked by surrounding factor, such as inflammationand promoted to excessive immune reaction, but it is not known which induceit, maybe continue stimulation of antigen or immune abdominal regulation. According to the theory of Traditional Chinese Medicine(TCM), diarrhea canbe induced by digestive disfunction of the spleen and the stomach, the smallintestine's disfunction of differing clear and turbid material, and largeintestine's failure to convey digest induced by irregular diet, exterior evils,abdominal affection and long-termed diseases. The main pathological place isthe spleen. And the syndrome of deficiency of spleen and stagnation of liverqi comes due to spleen's function suppressed by the liver. If the long-termeddisfunction of the spleen affected the kidney, appears the syndrome ofdeficiency both in the kidney and spleen. The symptom of bellyache comes ifstagnation of qi and stasis of blood is induced by deficiency of kidney andspleen, and abdominal mass forms with the development of disease. If interiorheat from stagnation of humidity hurts vessel, or if spleen qi is too deficientto control blood flow, hemafecia appears. In a word, the pathological mechanismof CD in TCM can be divided into exterior evils and interior hurts. There is few experimental report although curative effect of TCM on CDis significant, so this research is aimed to observe the curative effect andimmune mechanism of Kelong Qing infusion, the empirical formula in our hospital,on the rat with CD to provide treatment of TCM on CD with experimental evidence.2 Methods The model of CD is made on rat with Trinitrobenzenesulfonic Acid(TNBS)and alchohol, and the rats with CD were divided into four groups, namedrespectively as normal group, model group, Salicylazosulfapyridinegroup(SASP group) and Kelong Qing group. The normal group and model group weregiven water in intragastric way, and the SASP group and Kelong Qing group weregiven SASP and Kelong Qing infusion respectively in intragastric way. All thegroups were given once a day and continually for two weeks. And the curativeeffect and immune mechanism of Kelong Qing infusion on experimental rat withCD is explored according to the changes of modern medicinal index from entirety,parts and molecular levels. And all contents of this research includes follow: (1)General status, pathological changes and pathological changes undermicroscope were observed on the fifth day, on the tenth day and on the 15thday after being administered. (2) The disequilibrium of Thelper cell 1(Th1) and Thelper cell 2(Th2) werestudied through determining Interferon-γ(IFN-γ) and Interleukin 4(IL-4) inserum. (3) The cytokine before inflammation were studied through determining IFN-γand IL-4 in colon. (4) The factors of suppressing damage were studied through determiningNitric Oxide(NO) in serum and Myeloperoxidase(MPO) in colon tissue. (5) The factors of adhesion molecule were studied through determiningInterior Cellular Adhesion Molecules(ICAM)in colon tissue.3 Results (1) The ulcers and inflame manifestation in different degrees were observedon tissues of rats in model group based on general manifestations,pathomorphological changes and the level of MPO in colon tissue.
