There is the same ischemia/reprefusion injury of the brain in the acute stage of apoplexic stroke. During the stage, nerve cells, glial cells and microvascular endothelial cells are three sites of ischemia cascade reaction, researchers always show solicitude for them. About 90 percent cells in the center nerve system are the glial cells. And the most of them are the astrocytes, which are 10 times more than neuron. The astrocytes and their processes benefit for glia limitan, and keep the structure stabilization of blood vessel, neural soma, axon and synapse. The astrocytes join to make up of blood brain barrier, which are the first parts of central nerve system can be induced by the changes of the blood. In the pathologic stage of the brain ischemia/reprefusion injury, the normal cellular modality and function of astrocytes will change, and the astrocytes will release lots of nerve transmitters, nerve nutritional factors, celluar factors etc. and form glial scars by their enlargement and hyperplasia, and have the reciprocity with the neurons in the same pathologic stage. So astrocytes are very important in the acute stage of apoplexic stroke. After review the articles in lately five years, the research of astrocytes has made great process, but which is still little about effect of traditional Chinese medicine (TCM) on astrocytes in the pathologic stage. So this topic will do some work on it.This research project is therapeutic principle of "supporting health energy, strengthening body resistance and protecting brain" for the treatment in acute stage of apoplexic stroke, which is advanced by professor Sun sulun, in connection with that of "eliminating pathogenic factors" widely accepted in TCM to complete into integrated therapeutic system. Codonopsis pilosula(Franch.)Nannf. is the representative Chinese herb of the therapeutic method of "supporting health energy and supplementing Qi" in TCM. The research will study the extraction of herbs and effective components of Codonopsis pilosula(Franch.)Nannf. which effects on the astrocytes' ischemia/reprefusion injury. The cell survival rate was detected by 3-(4,5-dimethylthiazolyl) 2,5-diphenyltrazolium bromide(MTT)chromometry assay, necrosis and apoptosis were observed by double stain of Hoechst 33342 and propidium iodide(PI) with fluorescence microscope, lactate dehydrogenase (LDH) release was detected by chromometry. To observe the protective effect of Codonopsis pilosula(Franch.)Nannf. on the astrocytes' ischemia/reprefusion injury. In order to supply partial experimental foundation for the clinical therapy of the components of Codonopsis pilosula(Franch.)Nannf. and the therapeutic principle of "supporting health energy, strength'ening body resistance and protecting brain" for the treatment in acute stage of apoplexic stroke.The content of the experiment includes such methode:1.The toxicity of the components ofCodonopsis pilosula(Franch.)Nannf. to the astrocytes2.The double stain of Hoechst 33342 and propidium iodide(PI) with fluorescence microscope to detect the necrosis and apoptosis of the astrocytes on the hypoxia/hypoglycemia and reoxygenation injury3.The MTT assay to detect the survival rate of the astrocytes on the hypoxia /hypoglycemia and reoxygenation injury4.The chromometry of LDH to detect the LDH release rate of the astrocytes on the hypoxia/hypoglycemia injuryWe create a model of the hypoxia /hypoglycemia and reoxygenation of the astrocyte and certificated the components of Codonopsis pilosula(Franch.)Nannf. can protect it from the hypoxia /hypoglycemia and reoxygenation injury. We rudimently studied the protection mechanism of Codonopsis pilosula(Franch.)Nannf. and partial provide experimental foundation for the clinical therapy of the components of Codonopsis pilosula(Franch.)Nannf. and the therapeutic principle of "supporting health energy, strengthening body resistance and protecting brain".
