Objective: Parkinson's disease (PD) is a parapymmid progressive deteriorative disease in centre nerve system which always invade older people .Dopamine neurons apoptosis plays an important role in PD. Nowadays, acupuncture as a peculiar, safety, and convenient way has been applied in the treatment of PD and has good effection. The text will study the therapeutical effect and mechanism of the electroacupuncture(EA) on Parkinson's disease (PD), which will offer experimental basis for applying EA in clinic praction of the acupuncture -moxibustion. Methods: 90 Wistar rats were allocated into four groups by random: PD group, EA group, sham-operation group and normal control group. PD model was established by microinjecting 6-hydroxy-dopamine into mesencephalic substantia nigra in the right brain. The same amount of normal saline was injected into the rats of control group. After 4 weeks ,Apomorphine was injected into the rats abdominal cavity to induce rotation . The test were repeated every 7 days .Rats who had constant left rotation with a speed faster than 7r/min were selected as successful PD models.41 successful PD model rats were selected for treatment and randomly divided into two groups, model group (n=20) and electroacupuncture group (n=21). Acupoints of Fengfu and Taichong in EA group were punctured once a day for 14 days. The results were analysed by F-test and q-test.1 Observation on behaviour: 0.5 mg/kg of apomorphine was injected into abdominal cavity to induce left rotation. The number of circles rotated within 30 minutes after injection of apomorphine were recorded, twice a week for 2 weeks.2 Dopamine apoptosis content in substantia nigra: After 6% chlorahydrate ( CH) was injected to anesthetize the rats , the right substantia nigra was taken out and maked into specimen in 6μm every piece .Tyrosine hydroxylase(.TH)/TdT mediated dUTP-biotin Nick End Labeling(TUNEL) was carried out to judge the apoptosis of dopamine neurons, whose cell core should be dyed into purple.3 positive tyrosine hydroxylase (TH) neurons content in substantia nigra: The immune constitution chemical method was adopted to measure the positive TH neurons , whose cell plasm should be dyed into tan.4 Dopamine neurons content in corpus striatum :After the right corpus striatum was separated ,the wet weight was weighed by an analytical balance and added into 1ml 0.05mol/l HCIO4 It was centrifugated under 15000r/min for 30 min . Dopamine content was then assayed by hyper-potency liquid phase chromatograph ultravioletdector.5 superoxide dismutase(SOD) , glutathione peroxidase(GSH-Px) activity and glutathione(GSH) content : The right substantia nigra was seperated and weighed to form tissue supernate ,and then was taken after 15 min of centrifugation under 3000r/min. The immune constitution chemical method was adopted to measure the content of SOD, GSH-Px, GSH.All data were expressed in x ±s through SPSS11.0 software system. Result:1. In EA group, the number of rotations after the treatment was lower than before the treatment(P<0.05), but that didn't change in PD group(P>0.05).2. The dopamine content of the right corpus striatum in PD model was markedly lower than the normal group(P<0.01), and that in the EA group was higher than the PD group(P<0.05).3. The number of TH positive neuron on the right side in PD group was obviously lower than that of the normal group(P<0.01), and that in the EA group was higher than the PD group(P<0.05).4. In PD group, the number of apoptic dopaminergic neurons on the right side was obviously higher than those of the normal group(P<0.01). In the EA group, the number of apoptic dopaminergic neurons were lower than that of the PD group (P<0.05).5. The contents of glutathione peroxidase, superoxide dismutase and glutathione on the right substantia nigra in the PD group was lower than the normal group, and those in the EA group was higher than the PD group (P<0.01 and P<0.05).
