Xuezhikang On The Experimental Study Of The Protective Effect Of Kidney Of Diabetic Rats

ObjectiveTo investigate the protective function of Xuezhikang on kidney of diabetic rats and to discuss the possible mechanism.MethodsRemoved the right kidney from all the experimental rats. Divided the rats into three groups 2 weeks after the operation randomly: one group was uninephrectomized control group, two uninephrectomized groups were intraabdomino injected STZ and the blood glucose of the tail was tested 72 hours after injection. The one whose blood glucose was over 300mg/dl would be selected. Then divided them into two groups: one was diabetic group, the other was the group treated with Xuezhikang. At the end of 2nd week and 6th week after treatment, 6 rats of each group were sacrifized. Plasma glucose, serum cholesterol, trglyceride, LDL-C, urinary albumin of 24 hours , plasma albumin , plasma creatinine, urinary creatinine were tested, PAS staining was done on each renal section. We used image analyze system to measure the ratio of extracellular matrix to glomerular area and immunehistochemistry method to investigate the expression of TGF- P 1 and IV type collagen in renal tissues. All these data were analyzed by image analyze system.ResultsAt the end of 2 weeks after treatment, the urinary albumin of 24 hours , the excretion rate of albumin,TGF- 3 1 and IV type collagen were increased. Whereas the level plasma albumin was decreased in the diabetic rats, Xuezhikang could decrease the excretion of urinary albumin, increase the level of plasma albumin, and decrease TGF-3 1 and IV type collagen. At the end of 6 weeks after treatment, the serum cholesterol, trglyceride,LDL-C, urinary albumin of 24 hours,TGF- 3 1 and IV type collagen were increased, the Ccr was decreased, while the plasma albumin was decreased in the diabetic rats. Xuezhikang could decrease the level of cholesterok trglyceride, urinary albumin of 24 hours, the excretion rate of albumin.At thesame time,Xuezhikang could prohibit the decreasing of Ccr, and decrease TGF- 1 and IV type collagen .Conclusion 1 - Xuezhikang could correct the disorder of plasma lipid.2. Xuezhikang could decrease the excretion of urinary albumin and prohibit the decreasing of Ccr by regulation of cell fator.
