Network Through Experimental Diabetic Rats With Peripheral Neuropathy

Diabetic peripheral neuropathy is a continuous microvascular complication.As systemic complication of diabetes,recent evidence indicates that the retention of blood stasis in channels was involved in its etiology.Other reports also present evidence that significant amelioration can be provided by treatment with drugs of promoting blood circulation and dreging the channels.The experiment aimed at studying the important principle in the treatment of diabetic peripheral neuropathy.The etiology ,pathogenisis and clinical manifestation of retention of blood stasis in channels in diabetes mellitus,as well as the treatment of the principle,were summaried and generalized from the studies of documentary materials which can reflect the academic viewpoints of different schools of TCM.By means of experiment,the report tried to evaluate the protective effect and mechanism of Luo Tong on streptozotocin-induced diabetic peripheral neuropathy.The animal model of diabetes mellitus was set up by a single abdominal injection of streptozotocin(65mg.kg"') on rats. The powder of Luo Tong was mixed with the food(4g.kg"'.d"1).All experimented rats were killed 3 and 9 months.Hemodynamics triglyceride(TG) tissue plasminogen activetor(t-PA) and plasminogen activetor inhibitor(PAI) were observed.Thigh deep nerve preparations were observed with light microscope and transmission electronic microscope,some of them utilized Masson staining and immunohistochemical method. My el inated fibre density of Masson staining was counted.Neurofilament of immunohistochemical method was observed quantitatively. Substance P was quantitatively tested by computer. Reduced neurotransmitter was originator;after that was reduction of myelinated fibre density.Treatment with orally administered Luo Tong prevented essentially all of Substance P , myelinated fibre density abnormalities,and so were various indexs.Treatment of diabetic patients with Luo Tong may be helpful in delaying or possibly preventing the underlying cause of diabetic peripheral neuropathy,and this effect may be corrected the disturbance of hemorrheology,triglyceride,and plasma t-PA PAI activities.The composition of prescription of Luo Tong was made of Rhubarb rhizome > Peach kernel.
