Fever is a common symptom of all sorts of disease, especially some communicable disease, and is also one kind of the systemic reaction responses to infection and trauma.It is becomes the question which is urgent needs to solve at present that how reduces the fever course, alleviates the asthenia, inertia, immunosuppression osteoporosis and insomnia by the sick, and how precludes the ill effect of antiviral, antibiotics, and hormone. The Chinese traditional medicine such as herbs of qing re jie du have demonstrated its superiority and rationality in compound prescription increasingly when treating febrile disease compared with pure western medicine. Nao Re Qing Oral likuid (NRQ) is the effective Chinese traditional medicine in treating refractory fever after cerebral operation. To explore the antipyretic and detoxication mechanisms of NRQ, in this experiment the febrile model in rabbits were duplicated by endotoxin(ET) and edogennous pyrogen(EP), at first observed the influence of NRQ on body temperature in the febrile rabbits from the whole; second, detected the cytokine, pyrogenic and antipyretic medium in center; Simultaneously the immunosuppression model of mice were duplicated by hydrocortisone, observed regulation of the specific immunity function and non-specific immunity by NRQ; In vitro experiment the original generation of mouse peritoneal macrophage were used to observe the reactive oxygen species and free calcium ions by laser scanning confocal microscopy and the regulation of NRQ. In short, this experiment has explored the antipyretic and detoxication mechanisms of NRQ from the whole, the molecular, and the cell levels. The experiment has been divided into three parts. The first part: The antipyretic effect of NRQ and its mechanism. The febrile models of rabbits were established by ET and EP separately. The Aspirin (ASP) was taken as positive comparison. Firstly, observes the antipyretic effect of NRQ. (1) The NRQ group's maximum body temperature depth of camber(ΔT) and the bodytemperature response index (TRI) and the control group compares the not obvious difference(P > 0.05); (2) ET and EP group's ΔT and TRI remarkably are higher than the control group (P < 0.01 ); (3) NRQ and ASP treatment group animal body temperature rise peak value obviously is lower than ET and the EP group(P < 0.01), the antipyretic effect compared with the NRQ and ASP group, has not seen the obvious difference(P > 0.05). The result indicated 4 脑热清口服液清热解'机制的实验研究 that, NRQ has the obvious antipyretic effect on febrile rabbits induced by ET and EP. After obtains the antipyretic effect further discusses this medicine to the antipyretic mechanism, the result demonstrated: (1) NRQ have no effect on IL-1 beta cAMP and AVP in the hypothalamus (HP), the cerebrospinal fluid (CSF) and the Ventral septal area (VSA) of control group(P > 0.05). (2) NRQ and ASP can obviously reduce the IL-1β contents in HP of the ET and EP induced febrile rabbits(P <0.01). In addition, each group of IL-1β content and the body temperature change is being connected(r = 0.850,P <0.05). Which indicate that NRQ can remarkably reduce the febrile central cytokine. (3) The cAMP contents in the CSF and HP of NRQ and ASP treatment groups remarkably are lower than ET and EP group(P <0.01). The cAMP contents in the CSF and HP were paralleled with the changes of temperature separately also(HP:r = 0.940,P <0.01;CSF:r = 0.971,P <0.01). The results indicated that NRQ can decrease the content of central febrile medium. (4) The AVP content in VSA of NRQ and ASP treatment group compares with ET and the EP group separately, has the striking difference(P <0.01), the AVP content change and the body temperature change also presents the correlation(r = 0.777, P <0.05), the result indicated that NRQ can increase the release of central antipyretic medium.
