Object: Investigation on the effect and mechanism of incariin on the sexual activity of aged male rats.Methods: 20 months old spontaneous aged SD rats were separeted into 4 groups randomly: the blank control group, Andriol with Viagra control group, the high dose and the low dose of incariin group. Three weeks later, mating experiment was taken to observed the catching latency catching times mounting times the rate of catching and mounting. The prostatic and testicular ponderal indexes of all mice were measured. The testosterone index of rats' serum was determined using radioimmunoassay and the content of DA and 5-HT of rats' hypothalamus was measured with fluorospectrophotometry after hypothalamus homogenated. The content of smooth muscle and collagen fiber, the blood sinusoid lacuna of the corpus cavernosum of rats' penis, and the content of eNOS and ET-1 expressed in the corpus cavernosum of the rats' penis, stained with Masson' s and immuhistochemistry method respectixely, was measured under the optical microscope with Mias computer configuration image analysis system.Results: Incariin can improve sexual activity of senile rats evidently compared with controlled groups. It also can increase the content of DA and 5-HT of rats' hypothalamus as well as the contentof smooth muscle and eNOS of the corpus cavemosum of the rats' penis. Contemporarily, it can reduce the number of collagen fiber and the content of ET-1 of the corpus cavemosum of the rats' penis.Conclusion: Incariin can improve sexual activity of aged rats evidently. It can enhance the sexuality and reinforce erection of rats too. The results suggest that the effect mechanism of incariin have great relation not only to its increasing the male hormone level and the content and activity of monoamine neurotransmitter in rats ' hypothalamus but also to its improving the erectile tissue structure, increasing the content of eNOS and decreasing the content of ET-1 expressed in the corpus cavemosum of the rats' penis.
