| Objective: Set up the rat model to estimate the treatment of YuLeShu on depressive . Investigate the influence of YuLeShu on the depressive behavior , the monoarnine neurotransmitters and monoamine oxidase, and thr ought it to probe the anti-depressive mechanism of YuLeShu.Methods: Isolated-living condition and chronic mild unpredictable stress are applied successively to set up DEP rat model. DEP rat is assessed by Sue rose solution consumption test and Open-field test. They are used to estimate the change of interest, the level of spontaneous motor activities and behavior of DEP rat model. Fluorophotometry is applied to detect the monoarnine neur-otransmitters(NE,5-HT and DA) and MAO of the frontal-parietal cortexes.Eval-uating the efficiency of YuLeShu on the depressive behavioral tests and cerebral monoamine neurotransmitters and MAO of DEP rat model through statistics means.Results: The DEP rat model manifests significant changes: the decrease of sucrose solution consumption, the droping of Open-field behaviors, the drop ing of cerebral monoamine neurotransmitters and the lifting of MAO .Middle-dose , and high-dose groups of YuLeShu augment the consumption of sucrose solution, besides improves the Open-field behaviors of DEP rat significantly. Middle-dose and high-dose groups of YuLeShu increase the cerebral NE and 5-HT concentration and decrease the MAO concentration of DEP rat significantly.Conclusions:Isolated-living condition and mild unpredictable stress are applied successively to set up DEP rat model .YuLeShu can promote the depressive behaviors and monoamine neurotransmitters concentration, simultaneity demote the MAO cerebral concentration of DEP rat model .The result present a dose-dependent effect. The anti-depressive mechanism of YuLeShu mainly is due to increasing the cerebral NE and 5-HT concentration and decreasing the activity of the MAO in the brain. |
