Effects Of KangChanning Prescription On The Contents Of Monoamine Neurotransmitter And Behavior Of The Rats Model Of Parkinson's Disease

ObjectiveParkinson's disease (PD) is a neurodegenerative disease in nervous system, which trends to occur in middle and aged people. With the growing of aging population, prevalence of PD increased and influenced the work and daily life, which brought a big burden to society and families. The etiology of PD is still unknown. Professor TIAN created the KangChanning prescription based on abundant clinical experiences and association of former researches of literatures, which was beneficial to nourishing Liver and Kidney, replenishing Qi and Blood and relieving spasm by subduing liver-wind and has made good clinical effect. This subject replicated model of PD by injecting 6- hydroxyl dopamine into the substantia nigra of the rats with stereotaxic apparatus. We preliminary studied mechanism of the KangChanning to treating PD by observing changes of behavior of the rats and effects on the monoamine neurotransmitters.MethodThirty two male Sprague-Dawley (SD) rats were randomly divided into four groups among which each group contained eight rats:sham operated group, model group, Traditional Chinese medicine group and Madopar group. Then we replicated the PD model by injecting 6- hydroxyl dopamine into the PD model of rats with stereotaxic apparatus. The next was to replicate models by using one spot and two spots injected method respectively, then compared the rate of their success. An injection coordinate: the back of bregma 5.0 mm, the right of flank 2.0 mm, the under-part of back 8.1 mm, where was injected with 4μl. Two injection coordinate:(1) the back of bregma 4.4 mm, the right of flank 1.2 mm, the under-part of back 7.8 mm ;(2): the back of bregma 4.8 mm, the right of flank 1.0mm, the under-part of back 7.8 mm, where each one was injected with 2μl. Two weeks after replicating models, model rats were induced to rotate to the contralateral side by injecting apomorphine (APO) into abdominal cavity and this behavior would be recorded. Rats in accordance with quantitative standard could be used for models(>7 turn/min).Then treating the model rats, we administrated KangChanning prescription (Tianma, Baishaoyao, Guiban, Huangqi, Danggui, Houpu, Fuling, Shuanxiong, Jixueteng and Zhigancao) to the Traditional Chinese Medicine group with the dosage of 33.5 g/ kg·d; another dosage with Madopar was about 125 mg/kg·d. Both the Sham operated group and the model group were given equivalent quantity of distilled water. Model rats were induced to rotate to the contralateral side by injecting apomorphine (APO) into abdominal cavity and this behavior would be recorded two weeks after gastric perfusion. Then we compared the changes of rotation behavior of the model rats in each group to explore the mechanism of PD. Rats were sacrificed after 6 weeks routine feeding, we obtained striatal and put them into ultra low temperature to assay the contents of monoamine neurotransmitter of this rat model.Results1. The success ratio of model of two spots injected method group showed higher success ratio than one spot injected method group.2. Two weeks after models were replicated, model rats were induced to rotate with APO. The revolving turns of the rats were more than 7 turns/min among the models of PD, the Traditional Chinese Medicine group and the Madopar group. The rats in Sham operated group showed atypical rotation with the rates less than 7 turns/min.3. Revolving turns of rats in both the Traditional Chinese Medicine group and the Madopar group decreased after treatment with KangChanning and Madopar for 2 weeks (P<0.05). The revolving turns of rats in both the Traditional Chinese Medicine group and the Madopar group did not show statistical significance (P<0.05).4. The contents of monoamine neurotransmitters of rats in both the Traditional Chinese Medicine group and the Madopar group increased after treatment with KangChanning and Madopar for 2 weeks (P<0.05). The contents of DA, DOPAC, HVA, NE, 5-HT increased after therapy with KangChanning and Madopar, but there was no statistical significance (P>0.05).ConclusionsInjecting 6-OHDA into the substantia parts of the rats'brain could damage the striaturn system and decrease the contents of DA, which mimic the phenomenon of clinical pathology in PD, and could be used for PD model. It had been proven that this models had a stable, well-reduplicate characteristic as well as the high rate of success. Meanwhile, they were in accordance with the mechanism of pathology and the clinical traits of PD. Compared with model group, the revolving turns of the rats after administrating KangChanning in Chinese Traditional Medicine group decreased obviously and the content of monoamine neurotransmitter in the striaturn of the rats model of PD increased. The mechanism of these effects mentioned above might be related with protecting the substantia striaturn, enhancing antioxidant capability, retarding the death of cell and clearing free radicals.
