Rheum officinale Baill. is one of the three species recorded as official rhubarbs in Chinese pharmacopoeia. It is a famous traditional material and has been used for a along history as cathartic, astringent and hemostatic, promoter of biliary, gastric, pancreatic secretions and antibacterial. In recent years, many clinical and laboratory studies on the rhubarb in the treatment of Chronic Renal Failure (CRF) have been reported, and its mechanism also has been extensively studied. Focused on the biological activity and the chemical consitunents, we carried out following studies.The radix and rhizoma of Rheum officinale Baill. was extracted with 95%EtOH . After the extractive was shaken with EtOAc, the EtOAc layer was repeatedly chromatographed over silica gel and Sephadex LH-20 to give twelve compounds . Chemical structures for seven of them were characterized on the basis of their physiochemical and spectral data as I : (-) -epigallocatechin-3-o-gallate , II : chrysophanol , III: emodin, IV: aloe-emodin, V : gallic acid, VI: chrysophanol-8-o- 3 -D-glucopyanoside, VII: chrysophanol-1-o-β -D-glucopyanoside .In the preliminary pharmacological experiment , we administered the compound I orally to rats with renal failure.The effects of it on blood levels of urea nitrongen ,were investigated.These have established a experimental referrence for the clinical application and novel drug research on CRF.
