Gegen is the common used classical Chinese medicine and has being used to treat Type 2 diabetes by physicians of TCM in clinical. This thesis studies its extract's (GG) effects and mechanisms of action on insulin resistance, which is the key pathogenic parameter observed in the natural history of Type 2 diabetes, in animal and cell models induced by dexamethasone by means of contemporary pharmacology and molecular biology. The results showed as: (1)Gegen decreases fasting insulin (FI) and insulin resistance index (ERI) and increases insulin sensitive index (ISI) of rats of insulin resistance induced by dexamethasone significantly; (2)Gegen can improve glucose tolerance of rats of insulin resistance but not to alloxan indued diabetes mice; (3)Gegen increases glucose intake of 3T3-L1 adipocytes and improves the insulin resistance of 3T3-L1 adipocytes induced by dexamethasone significantly; (4)Gegen can activate peroxisome proliferator- activated receptor gamma (PPAR r), make it bind to PPAR response elements (PPRE); (5)Gegen can up-regulate mRNA expression of PPAR v ,aP2 and adiponectin. In conclusion, Gegen can improve insulin resistance and its roles of improving insulin resistance is related to activate peroxisome proliferator-activated receptor gamma and regulate the mRNA expression of genes of aP2 and adiponectin. The study illustrates the effects and mechanisms of action of Gegen on insulin resistance deeply and demonstrates its values of application in clinical for prevention and treatment of Type 2 diabetes at a more advanced point.
