Objective: To study the effect of Rhizoma typhonii extract (RTE) on T Lymphocytic leukemia Cell Lines CEM.Methods: By techniques of cell culture in vitro, T Lymphocytic leukemia Cell Lines CEM were treated by RTE in 500ug/ml 1000ug/ml 2000ug/ml 4000ug/ml concentrations for 24h 48hr 72hr 96hr respectively, and two control groups were set up including normal group,1.0umol/L Arsenic trioxide group. Cell morphology was observed by light microscopy electronic microscopy and survival rate was calculated. The cell DNA contents and cycle distribution were observed and analyzed by Flow-cytometry to measure the ratio of cell cycle distribution and apoptosis.Results: (1) All of the four concentrations (500ug/mL 1000ug/mh 2000ug/ml 4000ug/ml) of RTE could inhibit CEM cell growth characterized by apoptosis characterized by pyknosis, complete nucleic membrane, swollen mitochondria and extended ergastoplasm etc. The apoptosis body was shown at the same time. The ratio of apoptosis was 28.2% when treated with 4000ug/ml RTE for 48hr and the cells were inhibited in G1 phase, and cells in G1/G0 phase were significantly increased. (2) RTE showed a dose and time-dependent effect.Conclusion: It is found that RTE could significantly inhibit the growth of T lymphocytic leukemia cell lines and induce apoptosis.
