Resveratrol (3,5,4'-trihydroxy-trans-stilbene), a plant antibiotic produced in a wide variety of plants in response to injury, UV irradiation, and fungal infection, has been shown to inhibit cell growth of several types of cancer. Leukemia is a hematopoietic malignancy and one of the most common human cancers in our country. T-cell acute lymphoblastic leukemia/lymphoma (T-ALL) is an aggressive leukemia/lymphoma and presents a challenging medical problem. Here, we have investigated the effects of Resveratrol on the proliferation and apoptosis in T-cell lymphocytic leukemia (T-ALL) cells. These T-ALL cells were derived from model mice, diagnosed as T-ALL, with the T-cell specific deletion of the Pten tumor suppressor gene. The Pten tumor suppressor gene encodes a phosphatase that negatively regulates the phosphatidylinositol 3-kinase (PI3K) pathway activity. Loss of Pten function is common in many tumor types. Flow cytometric analysis demonstrated that the T-ALL cells are positive for CD4 and western blot analysis confirmed the absence of Pten expression. In this study, we treated T-ALL cells with Resveratrol which concentration ranging from 3.125μM, 6.25μM, 12.5μM, 25μM to 50μM for different duration as 6h, 12h, 24h, 36h and 48h. The result showed significant decrease in cell growth in dose- and time-dependent manners. In addition, Resveratrol treatment showed a remarkable increase in apoptosis. Investigation of the mechanisms of the Resveratrol's efforts on inhibiting T-ALL cell growth and inducing apoptosis indicated that pathway was PI3K/Akt independent. These results suggest that resveratrol may have potential as a novel therapeutic agent for T-ALL, however, futhur analysis of mechanism is needed.
