Objective:To investigate the effects of Vitamin- K1 on osteoporotic fracture healing in aged rats.Methods:The status of osteoporosis in 20-month-old male rats was confirmed by total body bone mineral density measurement.Fracture healing model was established by osteotomy with a wire saw and fixation with a Kirschner pin on left femur.64 model rats were randomized into two groups:32 for control group,and 32 for Vitamin-K1 group.Vitamin-K1 group was given intraperitoneal injection of Vitamin-K1 at dose of 15mg/kg/3d,while control group was given an equivalent volume of saline.The rats were scarified in 2,4,8 and 12 weeks after operation,the callus formation was monitored by histological and interleukin-6(IL-6),bone gla protein (BGP),alkaline phosphatase(ALP),calcium(Ca) in the blood-serum,the bone mineral density measurement of the callus and the right femur was taken,normal radiographs were taken at 12 weeks.The measurement data was expressed by mean±standard deviation((?)±s).The experimental groups and the control groups were compared with the number of matching design of quantitative information t-test,and P<0.05 was for the difference statistically significantResults:1.Vitamin-K1 produced a significant increase in serum BGP,Ca×P,ALP and. decrease in serum IL-6.2.Vitamin-K1 group had higher bone mineral density(P<0.05).3.The histomorphological analysis revealed that the process of cartilage forming and transforming were advanced in Vitamin-K1 group.4.Normal radiographs showed Vitamin-K1 group had more callus formation.Conclusion:1.Vitamin-K1 can.advance the osteoporotic fracture healing in aged rats.by increasing BGP,Ca,ALP in serum and.decreasing IL-6 in serum.2.During the healing process,vitaminK1 can accelerate endochondral bone formation and make increase the bone mineral density of bone callus and systemic bone.
