Preoperative Fluorouracil Intraperitoneal And Intravenous Chemotherapy Pharmacokinetic Study

Objective: To observe and study the fluorouracil (5 Fu) content in different tissues and liquids after IP and IV administration before surgery of the carcinoma of rectum.Methods: 55 cases of patients with rectal neoplasms were treated with 750mg/m3 5 Fu IP and IV quickly. HPLC was used to determine the content of 5 Fu in the peritoneal cavity, portal vein and systemic blood at 60～80 minutes and in the peritoneal and paratumorous lymphaden at 210～240 minutes after injection. Results: (1) High concentration of 5 Fu was found in arious tissues by IP,the concentration of 5 Fu in peritoneal was 4 folds of the group by IV. (2) The concentration of 5 Fu in the peritoneal cavity, portal vein and systemic blood by IP was 16 times, 4.4 times and 2.7 times of the groups by IV.Conclusion:IP chemotherapy of 5 Fu before surgery has a significant pharmacokinetic advantage over the convention IV routes in patients with carcinoma of rectum...
