| Objective: To explore the effect of wide-range concentration arsenic trioxide on the growth of human breast cancer cell line. Methods: MCF-7 cell was cultured with 0.5μmol/L~16μmol/L arsenic trioxide in vitro. MTT test was adopted to detect the growth suppression effect of arsenic trioxide in MCF-7 cell line. Cell morphology was studied by light microscope and electron microscope. Apoptosis ratio and cell cycle was analysised by flow cytometry. Agarose gel electrophoresis was used to get the message of the DNA fragment. Results: 0.5μmol/L~16μmol/L arsenic trioxide can inhibit the proliferation of the MCF-7 cell line, especially 2μmol/L~16μmol/L, and the effect was dose-and-time dependent,the IC50 of 96h is 3.0μmol/L. Cell morphology and flow cytometry revealed the growth suppression was caused by apoptosis rather than necrosis, meanwhile, the cell cycle can be arrest at G2-M stage. Conclusions: Arsenic trioxide can induce the apoptosis in MCF-7 cell line effectively, it may become a new anticancer agent for breast cancer in clinic. |
