INTRODUCTIONThe Carbon Monoxide Poisoning ( abbreviated as CMP) is a com-mon poisoning diseases in clinic, and the rate of the fatality and muti-lation is very high . So it is a serious threat for the health of the people . In order to explore the mechanism of CO poisoning and the treatment methods of CMP , many scholars have carried some studies with great achievement . But as far as now , there have been no reports as of the changes of IL - 1# and IL - 8 during CMP , and thus no reports on the studies on preventing and treating for the IL - 1 β and IL - 8 increase of CMP patients. In order to confirm the changes of the IL - 1# and IL -8 of the CMP patients as well as the relationship between them , and to further explore the mechanism of CMP as well as to find out a more ef-fective treatment. The author established a rat experiment mode of a-cute carbon monoxide poisoning (abbreviated as ACMP) and through the method of radio - immunity determined the IL - 1β and IL - 8 con-centration of the rat cerebrum cortex during ACMP and after methyl-prednisolong (abbreviated as MP )was administered.Materials and methods1. The experiment animals and groupingWistar rats altogether 50 , among which 23 females and 27 males ,and their bodies weights about 210g, They were divided into 4 groups . That is ; Group A , the blank control group. Group B. the carbon monoxide poisoning group ( abbreviated as CO). Group C, the nega-tive control group (treated with normal saline). Group D. treatmentgroup (treated with Methylprednisolong)2. The establishment of CO poisoning model.The poisoning device is a self - made five liter sealed poisoning container. When poisoning , put the rats into the poisoning container ,and let them silently inhale CO of 2500 -3500mg / m3for 40 - -60 minutes . and take 50 ml of specimens every ten minutes . Take the CO rapid testing tube and O2 rapid testing tube to test the CO concen-tration and O2 inside the container , so as to maintain the definite con-centration of CO , Meanwhile, the concentration of O2 was maintain at about 21% . Make 3 test during the whole poisoning procession.3. The treatment intervention and preparation of specimen for the homogenate of cerebrum.For group A , anesthetize and take about 500mg of the cerebrum cortex without poisoning at the time of 10 minutes , 3 hours and 6 hours . Grinding it with normal saline at the rate of 500mg: 1 ml . The operating should be done at the circumstance of 0?C . After centrifuga-tion ,they were stored in the - 20?C refrigerate . For group B, after poisoning ,take blood specimen in angular vein and test the carboxy-hemoglobin ( COHb ) , then isolated the cortex at the time of the 10 minutes ,3 hours and 6 hours post anesthetizing after the model estab-lished . For group C , after the model established , inject normal sa-line into the abdominal cavity . Take the cortex at the time of 3 hours and 6 hours post anesthetizing after the model established . For group D , after poisoning , inject MP into the abdominal cavity . Then isola-ted the cortex at the time of 10 minutes ,3 hours and 6 hours. After the specimen were collected , take the method of radio - immune to test .4. statisticsAll the data are express as mean SD and analysis of variance andq test were taken for testing.Results1. After acute CO poisoning ,the of IL -1β and IL - 8 values of the rats cerebrum cortex all increased at the time of 10 minutes ,3 hours and 6 hours. Compared with the control group , there was a sig-nificant difference ( p < 0.01)2. The release of IL - 1β and IL - 8 in the cerebrum cortex can be inhibited by Methylprednisolong when poisoning at the time of 3 hours and 6 hours.DiscussionThe acute carbon monoxide poisoning (abbreviated as CMP ) is a disease that seriously threatens the people's health , and is the main factor of poisoned death .
