ObjectiveTo study the protective effects of JinXiangDan on myocardial ischemia-reperfusion injury, the myocardial ischemia-reperfusion injury model of rat was set up to produce relative experimental evidences, which proved the mechanism and the theory of JinXiangDan protecting myocardial ischemia -reperfusion injury. Method1. Fifty healthy male SD rats, ranging from 180g to 220g, were divided into five groups at random: group I (pseudo operation ); group II (ischema-reperfusion operation); group III (ischemia-reperfusion with DiAoXinXueKang 54mg/kg.d); group IV (ischemia-reperfusion with JinXiangDan 1620mg/kg.d); group V (ischemia- reperfusion with JinXiangDan 540mg/kg.d); There were ten rats in each group.2. The rats in group I and II were fed with 0.9% saline, while the rats in group III , IV , V were fed with 2.7mg/ml DiAoXinXueKang Liquor, 81mg/ml JinXiangDan Liquor, 27 mg/ml JinXiangDan Liquor respectively. Each rat was fed with 20ml/kg one time a day for ten days.3. The model of myocardial ischemia-reperfusion injury was created with the ligation of left anterior descending coronary artery lasting 30 minutes and postischemic reperfusion persisted 60 minutes also.4. We observed the incidence of reperfusion arrhythmias and the rasing Z ST of ECG (lead II) after loosening ligature it. All rats were collected blood samples from carotid artery, taking serum apart, tested serum for detecting ( Creatine kinase ) CK and ( Lactic dehydrogenase ) LDH. Finally all rats were killed and took the myocardial tissues for detecting the activities both ( Superoxide dismutase ) SOD and ( Glutathione peroxidase ) GSH-Px, the level of ( Malodial -dehyde ) MDA. Two myocardial samples each group at random were took for observing their structures under a light-microscope.Result1. Compared with group I , the incidence of reperfusion arrhythmias, the rasing Z ST of ECG, the content level of MDA in myocardial tissues, the level of serum CK and LDH of group II were much higher(P<0.01), while the activities of SOD and GSH-Px in myocardial tissues of the same group were much lower (p<0.01).2. Compared with group II, the incidence of reperfusion arrhythmias, the rasing Z ST of ECG, the content level of MDA in myocardial tissues, the level of serum CK and LDH of group III, IVand V were much lower ( p<0.05 or 0.01), while the activities of SOD and GSH-Px in myocardial tissues of the same three groups were much higher (p<0.01).3. Compared with group III, the incidence of reperfusion arrhythmias and the rasing Z ST of ECG of groupIV weren't significantly different (p>0.05). The activities of SOD and GSH-Px in myocardial tissues were much higher (p<0.01),while the content level of MDA in myocardial tissues, the level of serum CK and LDH were much lower (p<0.05 or 0.01).4. Compared with group IV, the incidence of reperfusion arrhythmias and the rasing Z ST of EGG, the activity of SOD and the level of serum LDH of group V weren't significantly different (p>0.05), the content level of MDA and the level of serum CK were much higher (p<0.05), the activity of GSH-Px was much lower (p<0.05).5. Compared with group V, the incidence of reperfusion arrhythmias, the rasing E ST of ECG, the activity of GSH-Px, the content level of MDA ,the level of serum LDH of groupIII weren't significantly different (p>0.05), the activity of SOD in myocardial tissues much lower (p<0.05), the level of serum CK much higher (p<0.05).6. Oberved through a light-microscope, the myocardial structures of rats of group I was normal, while those of group II were the worst damaged among five groups. The damage of group III, IV and V was less than group II. ConclusionJinXiangDan possesses protective effects on experimental myocardial ischemia-reperfusion injury in rats. JinXiangDan is superior to DiAoXinXueKang, whose effect is directly proportional to dosage within certain range.
