Objective:To reproduce hepatic fibrosis model of rats and observe the effect of Kang Xian Ruan Can Chong Ji (KXRGCJ) in treatment of different grope of those rats , especially to observe the effect on matrix metalloproteinase-1 (MMP-1) , tissue inhibitors of metal-loproteinase-1 (TIMP-1) mRNA expression ,So as to study the mechanism of KXRGCJMethods:Experiment of treating hepatic fibrosis : From the beginning to make models, 90 wistar rats were randomly divided into 6 groups: normal control grope (10) , ultra structure grope (16) , colchicines grope (16) , and ECM (KXRGCJ) gropes (16 respectively) . Except the normal grope ,all of the others (the model gropes) were induced to hepatic fibrosis by CCl4 and administered respectively with water at the dose of 2ml/100g/d (the ultra structure grope) , colchicines at the dose of 1mg/100g/d (the colchicines grope) , ECM at the doses of 0.6g, 1.2g, 2. 4g/100g/d (the low, middle, high dose ECM grope) . At the end of the eighth week, all of the remnant rats were killed, Some parts of liver were taken which would be used in the experiment,Result:(1) MMP-1 expression: Compare with the ultra structure grope, the level of the normal grope is modestly lower, but the difference is not marked (P>0.05) , the levels of the gropes treated with medicine are markedly higher (P<0.05) ; Compare with the colchicines grope, the level of the low dose ECM grope is modestly higher, but the difference is not marked (P>0.05) , the levels of the middle and high dose ECM gropes are markedly increased (P<0.05) ; Compare with the middle dose grope, the level of the low dose ECM is markedly lower (P<0.05) , the level of the high dose ECM is modestly increased, but the difference is not marked (P>0. 05) .(2)TIMP-1 expression: the model gropes are significantly higher than that of the normal grope (P<0.01) ; Compare with the ultra structure grope, the levels of the gropes treated with medicine are markedly lower (P>0.05) ; Compare with the colchicines grope, the level of the low dose ECM grope is modestly lower, but the difference is not marked (P>0.05) , the levels of the middle and high dose ECM gropes are markedly decreased (P<0.05) ; Compare with the middledose grope, the level of the low dose ECM is markedly higher (P<0.05) , the level of the high dose ECM is modestly increased, but the difference is not marked (P>0.05) .(3) ultra structure grade: The model gropes are significant higher than that of the normal grope (P<0.01) ; Compare with the ultra structure grope, the level of colchicines grope is not different (P>0.05) , The levels of the low, middle, high dose ECM gropes are markedly lower (P<0.05) ; Compare with the colchicines grope, the level of the low dose ECM grope is not different (P>0.05) , The levels of the middle, high dose ECM gropes are markedly decreased (P<0.05) ; The level of the low dose ECM grope is markedly higher than that of the middle, high dose ECM grope (P<0.05) , The difference between the levels of the middle and high dose ECM gropes is not marked (P>0. 05) . Conclusion:(1) KXRGCJ can increase the level of the MMP-1 mRNA expression and decrease the level of the TIMP-1 mRNA expression markedly.(2) Within the proper range, the effect can be improved with the increase of the dose of KXRGCJ.(3) The effects of the middle and high dose KXRGCJ are better than that of colchicines.(4) The difference between the effects of the middle and high dose KXRGCJ gropes is not marked, but the rats administered with the high dose KXRGCJ are easier to die. So we can not increase the dose randomly. If every factor is thought about, the effect of the middle dose of KXRGCJ is best.
