| Fructus Mume is the unripe fruits of prunus mume (Sieb.) Sieb. et Zucc. in rosaceae family. Wu Mei tasts property and meridian. Its tropism was sour and astringent, enters the channels of Spleen, Lung and Large Intestine.lt has the effects of astringing the lung to stop cough, astringing the intestine to stop diarrhea, promotes the production of body fluid to quench thirst and regulating the stomach to relieve colic caused by acaris. It's often used as Astringent Materia Medica, indicated for chronic cough from deficiency of the lung, and indicated for chronic diarrhea and dysentery.There are two kinds of processed products, cleaned dark plum and charcoal of dark plum, to be used in clinic .In order to meet the objective request of the modernization of TCM, the quality of dark plum must be controlled with the normalized standard. This text has studied the method of quality controlling of two kinds of processed products. And according to the request of "the technological scheme" of the "tenth five-year project" subject the standardization study in processing of "30 sorts of TCM sliced medicinal herbs", we make out the quality standard of the two kinds of sliced medicinal herbs of Fructus Mume.HPLC method for the setting up of the chromatogram fingerprint of Fructus Mume was established. The mobile phase is water-methyl alcohol-phosphoric acid (97:3:0.2). The detector wavelength is 235nm. The flow is Iml/min. The method is simple, rapid and reliable. Through determining the absorbed quantity of pigment, we set up the quality controlling method of " frying dark plum to charcoal but remaining property". The experiment research proved this method to be reproducible, easy and feasible.Three methods (HPLC, electric potential titration, HPCE) are utilized to determine the content of the organic acid, and the results and methods are compared. The electric potential titration is the legal method from Chinese pharmacopoeia, but it can only determines the gross organic acid. HPCE is newly used, but the signal noise is large and the sensitivity is relatively low. So it has a bad repetitiveness. It is comparatively simple for HPLC to operate, and the reproducibility is good. Therefore HPLC is suitable to be used in organic acid content inspection. |
