Viral hepatitis B caused by HBV is a common infectious disease, according to the statistics of WHO, more than 300 millions HBsAg positive cases in the world. In our country, The number of asymtomatic carrier(AsC) have been more than 120 millions and a few of them will eventually develop to cirrhosis of liver and hepatocellular carcinoma. Chronic hepatitis B has been greatly threatening the human health and lives, at the same time, there is no cure for it.The compound of P.amarus which is composed of P.amarus, Astragalus membranaceus and Panaxnotoginseng has the effects, such as clearing away heat and toxic materials, soothing the liver and invigorating the spleen and activating the blood circulation to eliminating blood stasis. Experimental and clinical studies have demonstrated that it has the effects of decreasing aminotransferase, anti-virus and protection of hepatocytes.In order to explore and evaluate the effect, safety and mechanism of P.amarus of Fujian and Guangxi species and the compound of the P.amarus on the protection of hepatocytes against immunological induced liver injury in mice, we carried out an experimental study on the liver injury model caused by Bacillus Calmette-Guerin Vaccine(BCG) plus bacterial lipopolysaccharides(LPS).Each mouse was injected iv with 0.2ml of the BCG suspension(5* 107 viable bacilli) via tail vein, and 12 d later with 7.5ug LPS in 0.2ml of saline. The mice were killed and blood was collected 12 h after the LPS injection for bioassay of plasma transaminases and pathological examination of liver samples; Nffl mice were divided randomly into 5 or 6 groups: namely normal group, model group, Bifendate group and treatment group; Evaluation was done based on the serum ALT, AST, the weights and the histopathological change. The results showed that:l.The levels of serum ALT, AST remarkably increased. The liver and spleen of the experimental mice became swollen.2 P.amarus of Fujian species and the compound of the P.amarus could significantly reduce the elevated serum ALT, AST levels and reduce the weights of liver and spleen as well as improve the necrosis of the liver. This indicates that they have liver protective effects. No such effect was observed in those using P.amarus of Guangxi species.3 .Histopathological examination showed that hepatocytes were damaged severely. P.amarus of Fujian species and the compound of the P.amarus could improve the necrosis of the liver.4.1n the acute toxicity experiment, f.canarus of two species and its compound were safe in mice.The paper also discusses the mechanism of Chronic Hepatitis B from the viewpoints of neuroimmunoendocrine network(NEI).
