Traditional Chinese medicine Caulis Trachelospermi is the vine stem with leaves of Trachelospermum jasminoides (Lindl.) Lem., performing the function of dispelling wind and removing obstruction in the meridians, promoting blood flow and detumescence, and is used for treating arthritis and other diseases. So the HPLC fingerprints of Caulis Trachelospermi and its product, which was obtained after treated in the macroporous resin method, were established in this study, and 19 common peaks were acquired. The chromatographic condition definited in the end was: column: ZORBAX Eclipse XDB-C18 (4.6×150 mm, 5μm), column temperature 30℃; mobile phase: Gradient elution by methanol: water ((10 : 90) (0 min)– (30 : 70)(15 min)– (40 : 60)(40 min) - (60 : 40)(60 min)); flow rate: 0.8 mL·min-1 ; detection wavelength: 230 nm. The similarity of the fingerprint of the crude drug was: 0.943, 0.934, 0.850, 0.961, 0.904, 0.959, 0.905, 0.959, 0.961, 0.784, 0.923, 0.955, 0.744 and 0.813. The similarity of the fingerprint of the products obtained in macroporous resin method was: 0.967, 0.967, 0.976, 0.914, 0.945, 0.986, 0.944, 0.966, 0.973, 0.925, 0.958, 0.971, 0.685 and 0.796. Online qualitative analysis of the chemicals in the HPLC fingerprint was studied by using the LC/MS technique, and the chemical structure of 3 known chemical substance including nortracheloside, tracheloside, and trachelogenin were deduced. Especially an unknown peak, whose retention time (tR) was 30.7 min, was deduced probably as mataeresinol. At the same time the mass-spectrogram provided rich information on the molecular weight of unknown peaks in the HPLC chromatogram. Ultraviolet and spectroscopical quantitative analysis method was established in this study, and the total lignans in the samples of both the crude drugs and its products isolated by macroporous resin method were detected by the established method. The content of total lignans in the crude drugs was: 6.16 %, 5.47 %, 3.33 %, 5.44 %, 6.01 %, 6.07 %, 5.46 %, 5.55 %, 5.59 %, 4.66 %, 5.73 %, 5.68 %, 7.43 % and 7.52 %. The content of total lignans in the product was: 91.01 %, 84.46 %, 85.00 %, 88.50 %, 86.69 %, 90.62 %, 87.46 %, 95.80 %, 82.20 %, 91.77 %, 87.05 %, 81.67 %, 96.42 % and 97.08 %.
